Abstract
Objective Research on the total flavonoids from Radix Tetrastigmae (Sanyeqing),which Influences the expression of regulatory T cells(Treg) of LLC tumor bearing C57BL/6Mice, to explore the possible mechanism of regulate immune and reverse tumor immune escape function of Radix tetrastigme to tumor-bearing mice, and to provide experimental basis for the clinical use of drugs.Methods LLC tumor bearing C57BL/6Mice models were established by tumor homogenate inoculation method and were randomly divided into normal group, tumor-bearing saline group, high dose group of the total flavonoids from Radix Tetrastigmae, middle dose group of the total flavones from Radix Tetrastigmae, low dose group of the total flavonoids from Radix Tetrastigmae.24h after inoculated, all groups were began to administered orally except the the normal group, once a day.,14days later, all groups of animals were sacrificed. Stripped the tumor tissue and weighed to calculate the tumor inhibition rate of each group.The flow cytometry was used to detect the percentage of Treg cells,which from the mononuclear cells of peripheral blood and spleen lymphocytes, ANOVA and independent samples T-test methods were used to Conduct Statistical Analysis of the corresponding samples.Results①The tumor inhibition rate (TGS):The high and middle dose groups of the total flavonoids from Radix Tetrastigmae are:51.4%、1.3%, the low dose group has no inhibitory effect.②Treg ratio in the mononuclear cells of peripheral blood:Tumor-bearing saline group and low dose group of total flavones from Radix Tetrastigmae compare with the normal group are significantly different.There are significant differences between tumor-bearing saline group and high dose group of total flavones from Radix Tetrastigmae, there is statistical significanc.③Treg ratio in the mononuclear cells of spleen:high and low dose group of total flavonoids from Radix Tetrastigmae compare with tumor-bearing saline group, there are significant differences.Compare with high and low dose group of total flavones from Radix Tetrastigmae, there is statistical significanc.The same statistical significanc between middle and low dose group of total flavones from Radix Tetrastigmae.Conclusion①The high dose group of total flavones from Radix Tetrastigmae has most remarkable effect on inhibiting tumor.Low dose has no inhibitory effect on tumor growth and promotes the development of tumor.②The high dose group of total flavones from Radix Tetrastigmae can significantly reduce the percentage of Treg cells in mononuclear cells of peripheral blood and spleen in mice bearing Lewis lung cancer.③The total flavonoids from Radix Tetrastigmae has two-way regulating effect on tumor development.and the effect is related with dose.
