Abstract


Objective:To study the function and mechanism of regulating and controling that different virulence ofmycobacterium tuberculosis reacts at the stress of infected host macrophages.Methods:1RAW264.7were infected respectively bacterial suspension of the international standard virulentstrain of Mycobacterium tuberculosis H37Rv strains (H37Rv) and Bacillus Calmette-Guérin strains(BCG).To detect respectively activity of superoxide dismutase (SOD) and nitric oxide synthase (NOS) bycolorimetric method on the1th,6th,12th,24thhour after infection, and to determine the content ofmalondialdehyde(MDA) and nitric oxide(NO) of infected macrophages.2Cu/Zn-SOD,iNOS proteinexpression were examined by Western Blot Western blotting.3Cu/Zn-SOD mRNA and iNOS mRNAexpression were detected by RT-PCR.Results:1In the test detecting SOD and NOS activity and MDA and NO content by Chemical colorimetricmethod: After infection of BCG strains,the cellular activities of SOD and NOS as well as content of NOgradually increased whereas the content of MDA decreased in RAW264.7. However,after infection ofH37Rv strains,the cellular activities of SOD and NOS as well as content of NO gradually decreasedwhereas the content of MDA increased in RAW264.7. There is statistical significance compared with thecontrol group (P<0.05).2In Western Blot in test to detect the change of Cu/Zn-SOD,iNOS proteinexpression: After infection of BCG strains,the cellular activities of Cu/Zn-SOD Level express of graduallyincreased in RAW264.7.However, after infection of H37Rv strains,the cellular activities of Cu/Zn-SODLevel express of gradually decreased in RAW264.7. There is statistical significance compared with thecontrol group (P<0.05). After BCG strains,H37Rv strains,control group infected RAW246.7,there is nodifference in the expression of iNOS protein.To further study the different virulence of Mycobacteriumtuberculosis infection macrophage iNOS protein expression differences,we analyzed the expression ofiNOS mRNA.3In the test of RT-PCR to detect the change of Cu/Zn-SOD mRNA and iNOS mRNAexpression: After infection of BCG strains,the cellular activities of Cu/Zn-SOD mRNA and iNOS mRNALevel express of gradually increased in RAW264.7.However,after infection of H37Rv strains,the cellularactivities of Cu/Zn-SOD mRNA and iNOS mRNA Level express of gradually decreased inRAW264.7.There is statistical significance compared with the control group (P<0.05).Conclusions:1The stress of host macrophages infected with Mycobacterium tuberculosis associated withvirulence of Mycobacterium tuberculosis.2lowly virulent Mycobacterium tuberculosis strains inducedmacrophages stress.3highly virulent Mycobacterium tuberculosis strains inhibited macrophages stress.
