Abstract


Objective:To do the treatment base for further researching L-Asp potential clinic therapy value and its anti-AML mechanism,and then explore useful drugs to heal AML. Method:Present investigation use HL-60cell line as experiment and ALL cell line-Jurkat as positive control to detect their inhibition concentration50(IC50)after L-Asp dealing by CCK-8,then compare their IC50deciding whether have statistic difference.And then use Annexin-V-FITC/PI, Rh23to detect cell prophase apoptosis.After that comparing their apoptostic rate to estmate statistic value.and observing apoptosis shap by Hochst33258Fluorescent Dyes. ResuIt:L-Asp can inhibit HL-60groth obveriously,the IC50of HL-60is5U/ml and2.1U/ml respectively after24h and48h L-Asp treating and the Jrkat’s is4.5U/ml and1.7U/ml.Thers is no stastic difference between the IC50of HL-60and Jurkat through Student-t testing method.Present investigation demonstrate L-Asp has the effect of inducing HL-60cell line to apoptosis.Conclusion:L-Asp has apparent effect of growth inhibition and apoptosis to HL-60cell line,further more the effect is equal to the effect of Jurkat which was inhibited and to be induced apoptosis by L-Asp.L-Asp has effect of inducing tumor cell to apoptosis in its ant-AML mechanism and clinic value to treat AML.
