Objective:To explore the expression and significance of NF-κB、TNF-α and COX-2in chronic alcoholic liver injury rats.Methods:1. To induce the model of chronic alcoholic liver disease:Wistar rats (CL level) were randomly divided into four groups, including three groups as the model group (n=20) treated with different concentration of ethanol(6%,8%,12%), and one group as the control group(n=20) treated with0.9%sodium chloride. The rats with water deprivation but no fasting were killed at the end of the20th week. The blood was taken from the apex, separated the blood serum, and then liver tissue of rats would be fixed in10%neutral formalin.2. The serum alanine aminotransferase (ALT) and aspartate aminotransferase (AST) activities were detected by auto biochemistry instrument.3. The pathology detection of liver tissues:The sample of liver tissues were fixed in10%formalin, using conventional materials, embedded in paraffin and then made sections which stained by HE and immunohistochemistry to observe pathological changes of liver tissues and the expression of factor NF-κB,TNF-α,COX-2respectively.Results:1. Values of serum ALT and AST in rats:Compared with the control group, the values of serum ALT and AST were significantly increased in every model group and ALT/AST>2, obviously difference(P<0.05).2. Pathology detection:Results of HE stain:the structure of hepatic lobule are intact, no degeneration and necrosis in liver cell in the control group. Liver cells around central vein became different severity of swelling and formed a few empty lipocytes, liver sinus narrow, inflammatory cells infiltrated in every model group, and pathological changes of high concentration were a little more serious than low group.3. Results of immunohistochemistry stain:3.1Positive cells of NF-κB distributed around central vein principally. NF-κB expressed in all model groups.12%alcohol group compare with the control group, significant in statistic (P=0.020, P<0.05). Showed by table1.3.2Positive cells of COX-2distributed around central vein principally. COX-2expressed in all model groups.12%alcohol group compare with the control group, significant in statistic(P=0.044, P<0.05).3.3Positive cells of TNF-a distributed around central vein principally. TNF-a expressed in all model groups.12%alcohol group compare with the control group, significant in statistic(P=0.001, P<0.05).Conclusion:1. NF-κB, TNF-α and COX-2all involved in occurrence and progress of chronic alcoholic liver injury.2. The expression of TNF-a, COX-2and NF-κB exists relativity.
