Abstract


Objective:Aberration of cell proliferation is considered to be the maincause of renal clear cell carcinoma (ccRCC). SCIN is an important actin-binding protein. As a regulon of F-actin, multitude complex functions of SCIN have been found. In addition to its role in actin filament remodeling, SCIN has some roles in several cellular processes, including cell exocytosis, differentiation, migration and so on. SCIN also plays a pivotal role in tumorigenesis possiblly. The aim of this study is to confirm that SCIN would play an important role in proliferation of ccRCC cells, which would provide scientific basis for a further understanding of the molecular mechanisms in tumorigenesis of ccRCC.Methods:1. Expression of SCIN in nine ccRCC cell lines was measured by Westernblotting.2. Construction of SCIN-shRNA lentiviral vector.3.769-P cell line was transfected by SCIN-shRNA lentiviral vector, RT-PCR and Westernblotting were performed to detect the expressions of SCIN.4. Alterations of ccRCC cells biological behaviors were analyzed by MTS assay, cell growth curve experiment and flow cytometry cell cycle analysis.5. RT-PCR analyzed the target genes after knockdown of SCIN.Results:1. Westernblotting detected that SCIN was expressed in all three ccRCC cell lines, and769-P cell line was chosen as experimental model.2. RT-PCR and Westernblotting confirmed that SCIN-shRNA lentiviral vector could decrease the expression of SCIN.3. MTS assay and cell growth curve experiment showed that the proliferation of769-P cells were suppressed significantly, flow cytometry cell cycle analysis results revealed significant G2/M arrest in cell cycle in SCIN-knockdown769-P cell model.4. RT-PCR results revealed that CDK1was decreased after knockdown of SCIN.Conclusions:Aberrant SCIN might participate in the progression of ccRCC cell cycle mediated by CDK1, resulting the malignant behaves of ccRCC.
