Abstract


Progesterone plays an important role in the physiological function inwomen. In recent years, progesterone and its receptor have been payed moreand more attentions by scholars in male reproduction. It is believed thatprogesterone, which receptor mRNA exists in male sexual glands, binding to itsreceptor,can regulate spermatogenesis and improve the fertilization of sperm.while the sperm from the unexplained infertility exhibit a low fertility may dueto the deficient expression of the progesterone receptor and insensitive reactionto progesterone.It has not been reported whether the genetic polymorphism ofprogesterone receptor is involved in male reproduction at present. The studywill discuss the relationship between polymorphism of progesterone receptor and male infertility.Materials and methods:The genetic polymorphism of progesterone receptor in the28cases ofnormal sperm,50cases of oligospermia,35cases of asthenospermia,21casesof azoospermia,11cases of teratozoospermic were detected in the currentproject by PCR-RFLP. All the blood samples collected from patients in thereproductive center of The Second Hospital of Jilin University were kept in thetest tube with anticoagulants. The blood samples were grouped by the semenparameter according to the WHO semen routine inspection standards in1987.DNA were extracted from all the blood samples. Specific primers weredesigned according to the published human PR gene sequence(Gene ID:AY525610) by Oligo.7.0software. After the amplification and purification ofPCR products, the purified products were sent to Shanghai ShengGong tosequence, then, the genetic polymorphism and its relationship with maleinfertility would be analyzed. Result:The results showed that there exited G/A single nucleotide polymorphismin progesterone receptor sites+331, however, there only two patients witholigospermia were genotype of A/G, the rest of all were homozygous type G/G.Meanwhile,306bp insertion/deletion polymorphism were detected in the maleprogesterone receptor gene introns G, only two cases with oligospermia wereT1/T2hybrid type. There was no homozygous type in the rest of the testedsamples.Conclusion:1. G/A single nucleotide polymorphism in progesterone receptor site+331were detected with higher frequency of G;2. An insertion deletion polymorphism with the fragment of306bp isdetected in introns G with higher gene frequency of deletion.3. It was concluded that there was no significant correlation betweengenetic polymorphisms of progesterone receptor+331G/A and PROGINS and male infertility in our tested group.
