Abstract


Objective: To observe betaine homocysteine in the serum of rats with epilepsy,hippocampal gamma-aminobutyric acid, glutamic acid, and inhibitory nerve receptors:gamma-amino butyric acid receptors and glycine receptor, therebystudy the pathogenesis ofepilepsy and the mechanism of action of betaine.Methods: male Wistar rats were randomly divided into normal control group(intraperitoneal injection of saline,1.0ml saline), epilepsy group (intraperitoneal injectionpentetrazole,1.0ml saline), sodium valproate group (the intraperitoneal injectionpentetrazole, sodium valproate orally), betaine, low concentration group (intraperitonealinjection pentetrazole, betaine orally), the end of the experiment the the rat orbital bloodserum in the same type cysteamine acid content; rapidly at low temperatures the brain,ELISA method to detect changes in the gamma-aminobutyric acid, glutamic acid content;rabbits infected fluorescence and Western bloting of detection of hippocampal gamma-aminobutyric acid receptor, glycine receptor changes; liver tissue the HE stainingmorphology changes.Results: homocysteine content of the epilepsy group and the normal group wassignificantly lower (P <0.01), betaine content of low concentrations of homocysteine areassociated with epilepsy group was significantly lower (P <0.05); ELISA results show thatthe gamma-aminobutyric acid content of the epilepsy group compared with the control groupdecreased significantly (P <0.01). Betaine in concentrations of gamma-aminobutyric acidcontent of the epilepsy group was significantly higher (P <0.01); glutamic acid content of theepilepsy group and the normal group was significantly higher (P <0.01), the concentration ofbetaine low high school compared with the epilepsy group decreased significantly (P <0.01).Immunofluorescence and western bloting test results, compared to the epilepsy groupgamma-amino butyric acid receptor expression and normal control group significantlyreduced (P <0.01), betaine high, medium and low concentration group was significantlyhigher than the epilepsy group (P <0.01); compared to the the epilepsy group of glycinereceptor expression with normal control group was significantly lower (P <0.01), betainehigh, medium and low concentration group was significantly higher than the epilepsy group (P <0.01); HE staining, sodium valproate group of animal liver tissue were observed seenvacuolar degeneration (P <0.05).Conclusion: The betaine having preferably antiepileptic effects, and no liver toxicity.
