Objective:Intracranial aneurysm (IA) rupture is the main cause that leads tospontaneous subarachnoid hemorrhage. However, there is no theory in the fieldof IA research which at present could completely interpret the mechanism ofIA’s occurrence and rupture. Along with the technological advances ofmolecular biology and molecular genetics, miRNA, as a highly conservativenon-coding RNA which can participate in cell proliferation, differentiation,metabolism, apoptosis, and ontogeny through regulating gene expression, hasbeen a new approach to conduct research on mechanisms of disease. This studywas designed to employ molecular biological methods to study whether thereare significant difference genes miRNA and to explore the mechanism ofoccurrence, development and rupture of intracranial aneurysm wall throughstudying significant differences of miRNA.Methods:This study collects22aneurysmal wall specimens of relatively completeremoval and superficial temporal artery from the same patients as the controlspecimens after the treatment of supratentorial craniotomy, clipping ofaneurysm at the Second Department of Neurosurgery in the China–Japan UnionHospital of Jilin university on December1,2009to November1,2011period.Itrandomly selects from6ones from22aneurysmal wall specimens and6specimens of corresponding patient’s superficial temporal artery. Through RNAextraction and purification, RNA marking of samples, chip hybridization, chip scanning and the employment of the SBC analysis system for miRNAmicroarray data analysis of sample aneurysm wall and superficial temporalartery, it finally comes to the conclusion on Significant Difference GenemiRNA of Intracranial Aneurysm Wall ((P-value <0.05, Fold Change>2).Results:There exist three significant difference genes miRNA between Intracranialaneurysm wall and autologous control blood vessel (superficial temporalartery): hsa-miR-566, hsa-miR-99b*and hsa-miR-1180.Conclusion:1. The study verifies that there exist significant differences in miRNAexpression. The data analysis obtained three significant differences genesmiRNA: hsa-miR-566, hsa-miR-99b*, and hsa-miR-1180.2. Though related studies of hsa-miR-566, of hsa-miR-99b*, andhsa-miR-1180, inferences are drawn: hsa-miR-566is related to inflammatoryimmune responses; hsa-miR-99b*to TGF-β-mediated cell extracellular matrix;and hsa-miR-1180to cell apoptosis. Therefore, it proves the occurrence,development of intracranial aneurysm may be caused by the combined result ofseveral mechanisms: damage of inflammatory immune response, vascularremodeling caused by abnormal changes of the extracellular matrix, andover-express of vascular smooth muscle cells apoptosis.3. Whether hsa-miR-99b*is the specificity of the artery wall organizationexpress rise, whether can be used as the organization of the aneurysm genediagnosis index needs to be studied further.4. Atherosclerosis plays a very important role in the occurrence anddevelopment process of intracranial aneurysm, however it is uncertain on itsrelation with hsa-miR-566, hsa-miR-99b*and of hsa-miR-1180. Further research is needed.
