Abstract


Objective:To explore Ouabain reversing the drug resistance of human hepatoma cell in vitro and the possible mechanisms.Methods:1.Adriamycin-based discontinuous impact method with large dosage makes Bel-7402into adriamycin-resistant human-hepatoma-model which is Bel-7402/DAM cell strain.2.Real-time PCR detects Bel-7402and Bel-7402/DAM the mRNA expression level changes of MDR1before and after Ouabain intervention. Western-blot tests Bel-7402and Bel-7402/DAM the P-gp protein level changes before and after Ouabain intervention.3.MTT method detects influences of Ouabain on drug resistance of Bel-7402and Bel-7402/DAM cells.Results:1.The sensitivity of drug-resistant Bel-7402/DAM cell strain to adriamycin compare with non-drug resistant Bel-7402cell strain reducing obviously; and corresponding to this. multi-drug-resistant gene MDR1and the expression of its functional protein P-gp have been up-regulation.2.After72hours of Ouabain intervention in vitro, the multiple drug resistance gene MDR1of Bel-7402/DAM and Bel-7402group cells and the expression of its functional protein P-gp have been down regulation.3.MTT method detect:Ouabain can improve the sensitivity of Bel-7402DAM cells to adriamycin obviously; Namely certain extent reverse the drug resistance of Bel-7402/DAM cells.Conclusion:In vitro experiment Ouabain can partially reverse the drug resistance of hepatoma cells and the possible mechanism is related to decrease cell membrane of liver cancer MDR1and P-gp protein expression.
