Abstract


Background and objectThe most common malignant tumors of oral and maxillofacial were tongue cancer and carcinoma of the buccal mucosa, and organization source was come from epithelial tissue, especially was Squamous cell carcinoma in our country. Tongue squamous cell carcinoma (TSCC) is the most common malignant tumor of oral cancer, It usually grows very fast with higher degree of malignancy and invation; early cervical lymph node metastasis often occurs with high rate of transfer. At present the treatment of tongue cancer was comprehensive treatment is given priority to. Comprehensive treatment is often used to surgery, radiotherapy and chemotherapy treatments. Chemotherapy was often used to reduce postoperative recurrence and improve the long-term outcome. Preoperative chemotherapy to the reduction of the lesions, intraoperative and postoperative chemotherapy help prevent the spread of the lesions.Epidermal growth factor receptor (EGFR) is a transmembrane protein, it widely distributed in the human body each organization cell membrane. RGFR in most tumor cells have different degrees of expression and it has close relationship with the tumor differentiation, malignant degree and degree of infiltration. In recent years, research shows that, according to the treatment of EGFR has in some cancer treatment, especially squamous carcinoma treatment has achieved good results and have finished drug used in the clinical treatment of tumor. Based on the above research, we select43patients with pathologic diagnosis of tongue squamous carcinoma tissue samples, determine the EGFR expression. Use the epidermal growth factor receptor tyrosine kinase inhibitor(EGFR-TKI) treatment the Tca8113cells in vitror,research the function of the epidermal growth factor receptor in tongue squamous carcinoma treatment.Method43cases of tongue squamous cell carcinoma paraffin blocks were collected from Pathology department of Lanzhou University Second Hospital dating from January2006to July2011.All the clinical data of the cases were accurated and no use of corticosteroids and other immunosuppressive agents were recorded one month before operation according to their medical history. Tumor specimens are the initial issuance of the primary tumor without preoperative radiotherapy, chemotherapy or any other treatment. Expression of epidermal growth factor receptor was detected in43cases of tongue squamous cell carcinoma paraffin blocks and the Tca8113cell with immunohistochemistry SP method. Using different concentration of epidermal growth factor receptor tyrosine kinase inhibitor treatment the Tca8113cells in vitror24to72hours, to detect cell proliferation activity with MTT method, to observe apoptosis situation with Flow cytometry. Analysis the experiment data with the SPSS17.0statistical software, Data to (x±s) said and pathological grading (a=0.05).ResultsEpidermal growth factor receptor positivesataining for the performance of the tan or brown, background not coloring. And the main distribution in the cell membrane, a small amount of distribution in the cytoplasm.31(72.09%) cases of EGFR positive expression and12(27.91%) cases of EGFR negative expression in43patients with tongue squamous carcinoma.Tca8113cell tested positive for EGFR expression.Different concentrations of epidermal growth factor receptor tyrosine kinase inhibitor restrained the Tca8113cell proliferation,and the inhibitory action increased with increased of the drug concentration.The inhibition of epidermal growth factor receptor tyrosine kinase in the Tca8113cell at48h was obvious than the inhibition at24h and72h. The concentration gradient and the difference between the time point have the statistical significance (p<0.05).The apoptosis of the group of epidermal growth factor receptor tyrosine kinase inhibitor was more, about12.58%.the apoptosis of blank control group was few, about1.55%.The disparity of the two groups was obviously.Conclusion1The high level expression of epidermal growth factor receptor in tongue squamous cell carcinoma tissues prompt epidermal growth factor receptor play an important role in the process of tongue squamous carcinoma.2Epidermal growth factor receptor tyrosine kinase inhibitor restrained the Tca8113cell proliferation,and increase apoptosis rate of the Tca8113cell.
