Abstract


2009-2010 Xinjiang Shihezi, Manas, Hutubi County, Jimsar County, and Shuo County 15 processing tomato main producing areas in-depth investigation and found that in addition to the performance of field tomato mosaic, fern leaves Common symptoms of viral diseases, there are some plants showed all the yellow top, while the lower leaves appear normal, the symptoms are inconsistent with the ordinary yellow, field-like random distribution, the incidence rate of 2% -50%, between different regions incidence and disease severity have some differences. Morbidity and varieties, cultivation methods, and there is no obvious relationship between irrigation methods. By ICP-AES method of processing tomato yellow top elements 9 samples were determined, the results show: Top yellow plants Mg, Mn element content is slightly lower than normal plants, and B, K, Mo, Na, Zn of content was significantly higher than that of normal plants the same lot, Fe, Ca content of the element in the normal yellow plants plants were not significantly different. Samples of virus particles through the field of electron microscopy crude extracts and found a variety of virus particles present. Indirect enzyme-linked immunosorbent assay for the 30 field samples tested results indicate detection of CMV was 100%, TMV, ToMV, BBWV detection rates were 66.7%, 96.7%, 70.0%. The use of TMV, ToMV, CMV, PVS, PVM specific primers and other tomato yellow top samples by RT-PCR, to obtain the corresponding target band, which was consistent with the ELISA test, samples of tomato yellow top There are a variety of viruses exist. The PVM-specific primers PVMR / F target amplified fragments were cloned and sequenced, after the other with GENEBANK PVM reported partial sequence alignment, results showed: RT-PCR amplification of the target band is a section of PVM genes, there are 1198 nucleotides, including complete PVMCP gene (denoted as PVM-XJ). And other representatives of the gene pool isolates compared with the nucleotide sequence in Hangzhou, China (AJ437481), United States (AF023877), Poland (AY311394), Iran (EF397743), Latvia (GQ496609), Russia (NC001361), Germany ( X57440), Italy (X85114), and Canada (EF063383, EF063384, EF063387) homology region isolates 74.0% -95.8%. PVM-XJ with Russia's accession number NC001361 closest relationship, relations with Canada EF063387 furthest. For the first time in the processing tomato identified by molecular detection methods to PVM.
