Abstract


MrgC receptor (Mas-related G protein-coupled receptors) only specifically distributed in the dorsal root ganglia (dorsal root ganglia, DRG) and trigeminal ganglia (trigeminal ganglia, TG) of the small neurons, suggesting that these receptors may involved in information processing of pain or morphine analgesia play. Work in our laboratory showed that rats MrgC (Rat Mas-related G-protein coupled receptor C, rMrgC) can modulate morphine tolerance . In this study, tissue culture , SiRNA, quantitative PCR technology and other experimental method rMrgC SiRNA modulation mechanism of morphine tolerance . The results showed that , rMrgC SiRNA suppression caused by chronic morphine increased expression of CGRP mRNA and glutamate transporter (EAAC1) mRNA expression decreased . In the past, studies have demonstrated that after chronic morphine increased calcitonin gene- related peptide , glutamate transporter inhibition is an important reason leading morphine tolerance . The results of this study suggested , rMrgC may inhibit chronic morphine causes increased calcitonin gene-related peptide mRNA and glutamate transporter (EAAC1) mRNA decreased, keeping morphine analgesia effect.
