Abstract


The Linaria acid is a novel structure with chiral tricyclic quinazoline alkaloids , the thesis Liu wearing Research Development of fish acid investigate and optimize the synthetic route reported , Linaria acid synthesis and preliminary biological activity of other aspects . The willow wear according to the reported fish acid synthesis , we optimized synthesis process improvement . Recrystallization instead of column chromatography to separate the purified final product , the reaction yield was improved from 35% to 50%. In addition, we also fully synthetic route final step cyclization reaction involving the reaction reagent was investigated. Improved Linaria acid synthesis , we designed and synthesized 25 willow wear fish acid derivatives , are no reported compounds . Wherein , by nitration reaction succeeded in introducing nitro in Linaria acid benzene ring , ~ 1H-NMR and H, H-NOESY NMR Both methods identified the nitration reaction locus is Linaria acid C-7; the obtained 7 - nitro Linaria acid reduction to give 7 - amino Linaria acid . The other target compounds are used the corresponding substituted benzaldehyde as a starting material synthesis . All final products and intermediates of the chemical structure was confirmed via ~ 1H-NMR and MS , the final product was determined Specific rotation . Fish to wear one of the five compounds and Liu acid antioxidant activity of the initial screening results show that : LA -1 , LA -3 , LA -4 , LA -6 , LA -7 and the Linaria acid (LA) are can significantly reduce the lipid oxidation products MDA content in the liver tissue of aged rats , and a significant dose - dependent . LA-6 , the highest activity , its half of the inhibitory concentration 0.957nM . Other derivatives of the pharmacological activity of the test work is ongoing .
