To investigate the effects of Endocrine Disrupting Chemical Fenvalerate (FEN) on the reproductive performance in male chicken.24 male chickens were randomly divided into 3 groups. The chicken from each group were orally administrated with 0,10,50 mg/kg of FEN everyday for 4 weeks. The semen quality, testicular structure, blood gonadal hormone were examined after four weeks. On the forth week, semen was collected and inseminated the healthy hens through artificially insemination method, the fertilized eggs were collected to detect the fetility and hatchability.Additionally, in order to study the effect of FEN on the fertility in female chicken,16 laying hens were divided into control group (receive vehicle only) and treatment group which was orally administrated with 10 mg/kg FEN everyday for 6 weeks. After 3 weeks treatment, the eggs were collected and the egg quality was detected, after 5 weeks, hens were bred with the semen from healthy male chicken through artificial insemination method, afterwards the fertilized eggs were collected to detect the fetility and hatchability.1. Effects of FEN on the reproductive performance in male chicken(1) Semen qualityOur results showed that the FEN had adverse effect on semen quality. The result showed that sperm motility from two treatment groups was significantly decreased, while the spermic abnormality rate were significantly increased compare with control group (P< 0.01).(2) Testicular morphologyThe results of testicular morphology showed that the diameter of semeniferous tubuli, the thickness of spermatogenic epithelium, the tuble area and the lumen opening were significantly reduced in 10 mg/kg treatment group(P< 0.05), the area of semeniferous tuble was significantly reduced in 50 mg/kg treatment group (P<0.05), while the diameter of semeniferous tubule, the thickness of spermatogenic epithelium and the lumen opening were not changed in 50 mg/kg treatment group.(3) Serum T and E2 The serum hormone results showed that FEN significantly reduced the level of serum E2 (P< 0.05), while no effect on serum T in all treatments.(4) The rate of fertilized egg and hatchabilityAfter 4 weeks treatment showed that, the percentage of fertilized egg was lower in the treatment of 10 mg/kg FEN as compared with the control group, it was 83.87% in control, but 74.63% in 10 mg/kg FEN. The hatchability was lower in the treatment of 10 mg/kg FEN than that of the control, it was 80.65% in control, but 67.16% in 10 mg/kg FEN.The percentage of fertilized egg was lower in the treatment of 50 mg/kg FEN as compared with the control group, it was 73.53% in control, but 65.12% in 50 mg/kg FEN. The hatchability was lower in the treatment of 50 mg/kg FEN than that of the control, it was 70.59% in control, but 64.18% in 50 mg/kg FEN.2. Effects of FEN on the reproductive capacity in female chicken(1) Egg qualityAfter 3 weeks treatment of 10 mg/kg FEN on hens, the albumen height of egg, the color of egg yolk, and the haugh unit were not changed, but FEN treatment significantly reduced the thickness and the intensity of eggshell (P<0.05).10mg/kg FEN have slightly effect on the rate of egg fertilization, the egg fertilization rate was 76.9% and 75.75% in control and treatment respectively. However,10 mg/kg FEN obviously reduced the hatchability, which was 73.08% in the control, but 48.50% in the treatment.Taken together these data suggest that:(1) FEN has adverse effect on the fertility in adult male chicken, it can damge the histologic structure of testis, decrease the area, smaller seminiferous epithelium and sperm motility, increase sperm abnormality, reduce the rate of egg fertilization and hatchability in mated hens. (2) FEN inhibited the secretion of estrogen and disrupted the ratio of E2/T, which may lead to cock breeding performance decline. (3) FEN decrease the thickness and intensity of egg shell, decrease the hatchbility of fertilized egg
