Abstract


Objective To investigate the cysteine ??aspartate specific protease the -1 (caspase-1) and its active cytokines interleukin -1β (IL-1β) expression after acute spinal cord injury in rats and its significance . Methods Adult male Sprague-Dawley (SD) rats were the 144 randomly divided into the control group and the spinal cord injury group ( n = 72 ) . Two groups based on injury drawn at different times each subdivided into 12 subgroups . Spinal cord injury group using a modified Allen's heavy objects against established spinal cord injury model ( with a diameter of 2.5mm, 10.0g quality stainless steel rods along with graduated glass pipe from 25mm height vertical free whereabouts , impact T9 dural sac , resulting in rats with acute spinal cord injury ) , the control group only laminectomy . In the corresponding time points after surgery , the rats were sacrificed , drawn to the site of injury as the center , HE staining of pathological changes in the spinal cord , immunohistochemical staining of Caspase- 1 and IL -1β expression . Caspase-1 activated cytokines of IL- 1β in the control group rat spinal cord tissue has a small amount of expression ; 30min, caspase-1 expression in the spinal cord injury group and the control group after spinal cord injury there is a significant difference after spinal cord injury group expression gradually increased after injury on day 3 table peaked , then decreased gradually to 2 weeks spinal cord injury group and the control group, the expression of non- significant with differences ; 30 min after spinal cord injury of IL - 1β the expression of spinal cord injury group and the control group significant difference after spinal cord injury group expression increased quickly , peaked at 6 hours after the injury table , and then decreased gradually to 2 weeks of spinal cord injury group and the control group was no significant difference . The conclusions normal memory in a small amount of caspase -1 , IL -1β expression in rat spinal cord tissue ; acute spinal cord injury in rats after 1 week of caspase- 1 and IL-1β expression in spinal cord injury group and the control group, there were significant differences , Tip of caspase- 1 , IL -1β may be involved in secondary spinal cord injury ; 2-4 weeks after acute spinal cord injury in rats , caspase -1 and IL-1β expression in the spinal cord injury group and the control group no significant differences , suggesting that The biological role of caspase-1 and IL-1β on spinal cord tissue .
