Objective: To examine whether XNJI can be used for the treatment of cerebrovascular ischemia-induced lupus encephalopathy (CNS-SLE), and analysis of its mechanism of action. Methods: SLE model mice were divided into the model group XNJI treatment group, prednisone combined with cyclophosphamide treatment group (XNJI of 3ml / d prednisone 1mg/kg.d combined with cyclophosphamide amide 2mg/kg.d, an abdominal injection administered two weeks, the gap of two weeks); 4 months after the intervention, the drug using MRI head scan, analysis of the brain, the cerebellar hemispheres deep white matter, basal ganglia and brainstem abnormalities, and cerebrospinal fluid levels CNS-SLE mice learning and memory behavior determination step down; collecting serum drug intervention in the medium term (2 months) and late (4 months), application of enzyme-linked immunosorbent assay for detection of serum cytokines TNF-α, IFN- the γ; drug intervention four months later, the mice were killed, taking CNS-SLE mice brain tissue for pathological analysis, immunohistochemical analysis of VCAM1, Fas / FasL expression of P38 protein expression and phosphorylation levels using Real time RT- PCR (SYBR dye method) detect brain tissue vascular cell adhesion molecule -1 (VCAM-1) mRNA expression levels. Results: four months after drug intervention reduce the traditional Chinese medicine, Western medicine treatment group relative to the the model group limbs obvious erythema symptoms; according to MRI mapping analysis found that brain memory model group abnormal signal lesions and bleeding, the symptoms of traditional Chinese medicine and western medicine group slowed, cerebrospinal fluid content no significant difference in the three groups; reaction time, the number of errors in the three groups of the learning ability of rats was no significant difference (P gt; 0.05), memory retention ability treatment group jumped off the platform of the incubation period, compared with model group significantly increased (P lt ; 0.05), the number of errors no significant difference (P gt; 0.05); mouse serum of IFN-γ of TNF-α levels in the same time period no significant difference (P gt; 0.05), but the model group and the western medicine group final serum IFN-y levels were significantly lower than the medium-term (P lt; 0.05). Immunohistochemistry pathologic images show the presence of a large number of inflammatory cytokines in the brain tissue of the model group, bleeding points, edema, neuronal cell hyperplasia; such as traditional Chinese medicine and western medicine group and pathological features greatly reduce or disappear, neurons active proliferation; of the results of VCAM-1 and phosphorylation of P38 in traditional Chinese medicine and western medicine group brain tissue expression level significantly significantly lower than the model group (P lt; 0.05); FAS of FASL P38 in the three groups of brain tissue expression level without significantly with differences (P gt; 0.05); VCAM-1 mRNA expression levels in the three groups of mice brain tissue model group and there was no significant difference, while the model group and the western medicine group differences. Conclusion: Xingnaojing intervention effective in improving the symptoms of SLE model mice brain hemorrhage, inflammation, edema, enhanced memory ability of mice; The mechanism results suggest Xingnaojing intervention can reduce the secretion of IFN-γ expression in mice , inhibition of brain tissue of VCAM-1 and phosphorylation of p38 protein expression levels, thus affecting the process of an SLE model mouse secondary lupus encephalopathy.
