Synthesis of Coumarin Fluorescent Probes and Their Application for the Detection of Hydrogen Sulfide in Biological System

Abstract


Recently scientific studies have shown that H2S is one of the three gas signal substance in human body. H2S plays an important role in regulating physiological function, such as the reduction of nitroso and hemoglobin.Literature reported copper ions can cause the fluorescence quenching of the probe which containing carboxyl and nitrogenous heterocyclic. It is well known that H2S and copper ions has a strong complexation ability. When the quenched fluorescent probe and H2S react with each other, copper ions will coordinate with H2S and be separated from the probe then the probe fluorescence will recover. So we can detect H2S in this method.The probe employ the7-(diethylamine)coumarin-3-carboxylate as a fluorophore, and equipped with L-Asp and complex with the copper ion. We characterized it with MS, NMR and make cell experiment. The reaction completed in20s and the fluorescence recover. The fluorescence intensity and H2S concentration showed a linear relationship between the concentration of0.5-10μmol L-1. The detection limit is500nmol L-1. The probe is selective to hydrogen sulfide over cystein or other reactive compounds. The probe has high sensitivity and selectivity and responses quickly to H2S. At the same time it can be completely dissolved in water which is important for the detection of hydrogen sulfide in biological system.
