Abstract


The grass carp is one of the traditional freshwater aquaculture species of Chinese. Due to the big breeding density, much artificial diet, environmental degradation, inappropriate medication, hepatobiliary disease of grass carp has become the major diseases of the grass carp all over the China in recent years. Therefore, development of safer drugs for the treatment of hepatobiliary diseases of grass carp has a very important significance. Studies have shown that turmeric has anti-tumor, anti-oxidation, anti-inflammatory, lowering blood pressure, lowering blood pressure, protect liver function and other pharmacological effects. It can protect the mammalian liver which damaged caused by exogenous substances effectively. Therefore, the study of turmeric extract in the treatment of hepatobiliary disease of grass carp has a very important significance. This topic researched on optimizing the alcohol extracting process for turmeric, formulations of the Turmeric extract and the quality standards of the turmeric extract powder, has Laid the foundation for the development of traditional Chinese medicine treatment of grass carp hepatobiliary disease drug.The main contents of the subject and the the results are as follows:1. Research on the optimal alcohol extracting technology for turmeric:The extraction alcohol concentration, the extraction alcohol consumption, the extraction time and the number of extraction on effect of extraction volume of Curcumin, Curcuminoids and ointment collecting rate which were evaluated by orthogonal design L9(34). The results show that the best extraction process was every sample underwent3times extraction, each time adding10times the amount of80%alcohol and extracting for1.5hours.2. Research on the formulation Process of turmeric extract powder: According to the physical properties of the turmeric extract and the grass carp feeding habits, research on the formulation Process of turmeric extract powder. This research studied on the selecting accessories, drying method, drying time and the materials ratio of the turmeric extract powder, and then determined the best forming techniques are as following:The corn starch was Selected as turmeric extract powder’s excipient, the optimum dosage was the crude drug than accessories1:8. oven drying was used and the drying temperature was80°C, the drying time was10h.3. Research on quality control of turmeric extract powder:curcumin was used as standard sample, and TLC was used for the identification of turmeric extract powder. The result show that the optimal alcohol extracting technology for turmeric had fine separability and good durable ability. Then HPLC was used for the quantitative research of turmeric extract powder with the curcumin as standard sample. The results show that the extraction method has a good linear relationship, good accuracy, good stability, good reproducibility and recovery qualified. Three batches which were prepared by our laboratory were measured, the level of the contents of curcumin should not less than1.3mg.4. Preliminary pharmacological research of Anti-grass carp liver damage by using turmeric extract powder:turmeric extract powder was of low toxicity which Seen by the acute toxicity test of turmeric extract powder, The study of prevention and treatment of grass carp liver injury with turmeric extract powder show that turmeric extract powder is possible to improve the antioxidant ability of the grass carp liver, and it can repair grass carp impaired liver tissue, it has a good hepatoprotective effect.In summary, the optimal alcohol extracting technology with high extraction efficiency of curcumin and Curcuminoids for turmeric (the best extraction process was every sample underwent3times extraction, each time adding10times the amount of80%alcohol and extracting for1.5hours.) is stable. The design of formulation Process of turmeric extract powder (The corn starch was Selected as turmeric extract powder’s excipient, the optimum dosage was the crude drug than accessories1:8. oven drying was used and the drying temperature was80°C, the drying time was10h.)was simple and reasonable, it can be used for high-volume production. The quality control of turmeric extract powder with the curcumin as standard sample was stable and controllable. The pharmacology experiment proved that turmeric extract powder was of low toxicity and the turmeric extract powder is possible to improve the antioxidant ability of the grass carp liver, and it can repair grass carp impaired liver tissue, it has a good hepatoprotective effect. The accumulated experience and test data from the Institute has provided a reference for future new drug research and development of traditional Chinese medicine treatment of hepatobiliary disease of grass carp, and laid the foundation for the development of the follow-up study with turmeric extract powder.
