Phospholipid(PL), ingested as a dietary supplement, has therapeutical use as dietetic support in cases of organic debilitation and regulated fat metabolism. In addition, PL acts as a vehicle for essential fatty acids useful as growth factors, in the prevention of atherosclerosis and to protect against hypertension, thrombosis and ulcers. The Phospholipids mainly comes from the plant oils, as the PL level in crude soybean oils is approximately0.1~1.8%. However, the presence of PLs in the specific food may have various effects on the process and quality of the corresponding food. In oils, phospholipids are believed to contribute to the antioxidant properties of oils.They are also responsible for losses in neutral lipids in oil processing during neutralization, and contribute to the discoloration of the oil during deodorization and steam distillation. Currently, there are many methods for the determination of phospholipids, the national standard is the malachite green method However, the method have a variety of shortcomings, for instance require tedious procedure of sample preparation, time-consuming, and lower detection limits.In recent years, with the development of chemometrics, the fourier transform infrared spectroscopy (FTIR) has found widely application in the quantitative analysis of oils and fats. This paper established two ways of rapid analysis of phospholipid content in edible oils by using Fourier transform infrared spectrometer.The first method is a FTIR method for the determination of total phospholipids(PL) in edible oils has been developed based on the extraction of PL into methanol/acetonitrile solution. The spectra of sample extracts ratio against that of degummed oil extracts and the gap-segment2nd derivative processed spectra of differential spectra obtained were used to determination of PL. Among solvent investigated, methanol was optimal extractant. Addition of acetonitrile to methanol could improve the specificity for PL extraction. The band selection for PL quantization suggested that1243cm-1was superior to1740,1172,1090,970cm-1band. By measuring absorption at1243cm-1of asymmetric phosphate diester (PO2-) stretching in2nd derivative spectra, spectral interference from extracted triglyceride constituents were minimized. Using mixed solution of methanol and acetonitrile with the volume ratio of4:6as solvent to extract PL from edible oils and maintain the ratio of solvent and oil is2:1will contribute to the improvement of accuracy and precision in FTIR method developed according to comprehensive standard addition experiments and validation experiments of practical samples in comparison with Barlett arseno-molybdate method. Compare with Molybdenum blue method, the relative standard deviation for developed method is from2.45-3.88%, mean3.17%, the mean relative error is2.17%, showed good accuracy of models. It is promising for FTIR-PLS method to be used as a substitute for tradition wet chemical analytical methods for PL determination in edible oils.The second method is a rapid reliable method was developed to determine the total phospholipids content in vegetable oils by Fourier transform-mid infrared spectroscopy (FT-MIR) and partial least-square regression (PLS). PLS regression was employed to extract relevant information from the complex MIR spectra of phospholipids. Spectra preprocessing methods including standard normalization of vector (SNV), smooth and derivative were compared and finding that SNV corrected spectra gave the improved prediction performance over original spectra. Results suggested the bands between970cm-1-1320cm-1produced the better models than full spectrum PLS, lower RMSEC (316) and RMSEP(386) values as well as higher determination coefficients (0.9979) for oil samples with PL concentration ranging from200to20000mg/Kg. Compare with Molybdenum blue method, the relative standard deviation for developed method is from1.42%-3.52%, mean2.36%, the mean relative error is0.75%, showed good accuracy of models. It is promising for FTIR-PLS method to be used as a substitute for tradition wet chemical analytical methods for PL determination in edible oils.To sum up, this paper established two kinds of fast analysis of phospholipids in oil by using Fourier transform infrared spectrometer,the two methods are both simple and convenient, suitable for the detection of phospholipid content.
