Experimental Studies on Inhibitory Effect of Dihydroartemisinin Combined with Octreotide on the Growth of Human Cholangiocarcinoma in Vivo

Abstract


Objective: To study dihydroartemisinin (DHA) in combination with octreotide (OCT) treatment of human cholangiocarcinoma QBC939 nude mice xenografts role and explore its possible mechanism of action . Methods: human cholangiocarcinoma QBC939 nude mice tumor animal model 30 , were randomly divided into 5 groups : the Octreotide group, (100μg/kg), dihydroartemisinin group (80mg/kg), octreotide dihydroartemisinin combination group (50μg/kg 40mg/kg), blank control group (saline) , solvent control group ( vegetable oil ) . Each group were administered daily for 4 weeks , observed each group planting tumor growth , the mice were sacrificed 24 hours after the last dose , to complete stripping tumor block , weighed the bare tumor weight , calculated inhibitory rate ; using immunohistochemical methods were used to detect the expression of HIF-1α in tumor tissue and microvessel density ( MVD ) ; application TUNEL assay apoptosis of tumor tissue . Results : 1.OCT , DHA group implanted tumor volume and weight of the combined group was significantly less than the control group ( P lt; 0.05 ) , the implanted tumor volume and weight of the combined group was significantly less than the single drug group and the control group ( P lt; 0.05); 2. combined treatment group tumor inhibition rate Ea b = 58.67% , the DHA group of tumor inhibition rate was Ea = 29.20% the OCT group tumor inhibition rate Eb = 30.40% . The tumor inhibition rate of the combination group was significantly greater than the medication alone group (P lt; 0.05); calculated according to of Guinness the formula q = Ea b / (Ea Eb-EaEb) q = 1.16 gt; 1.15. 3. HIF-1α in each experimental group . expression is less than the control group ( P lt; 0.05 ) ; combined group of HIF-1α expression is less than each drug alone group ( P lt ; 0.05 ) , the DHA group and OCT group of HIF-1α expression differences ; 4. MVD of each experimental group than the control group ( P lt; 0.05 ) , which jointly MVD less than each drug alone group ( P lt ; 0.05 ) , DHA group and OCT group between MVD values ??no difference ; 5 in each experimental group tumor apoptosis index (AI) was significantly greater than the control group ( P lt; 0.05 ) , which combined group AI than medication alone group ( P lt; 0.05 ) and no significant difference in the OCT group and DHA group AI . Conclusion: dihydroartemisinin with octreotide in human cholangiocarcinoma in nude mice implanted tumor growth inhibition , the inhibitory effect is more pronounced when the combination of the two drugs , the mechanism may be associated with the induction of tumor cell apoptosis and inhibition of tumor angiogenesis .
