Pulmonary fibrosis (the Pulmonary fibrosis PF) is a disease of unknown etiology, pathology is characterized by early diffuse alveolitis, alveolar epithelial damage and the late massive pulmonary pathological proliferation of fibroblasts and collagen matrix progressive accumulation and replace normal lung organizational structure. Epidemiological survey found that this disease from the onset of symptoms to death in an average of 2-4 years, 5-year survival rate was 30% -50%. The pathogenesis of pulmonary fibrosis has not been completely elucidated, lack of effective treatment, the clinical still glucocorticoids and immunosuppressive agents and cytotoxic drugs mainly, but the effect is not ideal, and has significant side effects . Therefore, actively seeking a high efficacy, fewer side effects drugs is an urgent problem to be solved. Clinical, traditional Chinese medicine in the prevention and treatment of pulmonary fibrosis have a bright future, and has made some progress in clinical practice. Loquat Rosaceae loquat loquat leaves of bitter, slightly cold, lungs, stomach, with Qingfeizhike, Jiangni antiemetic effect. As a traditional medicinal plant, has a good anti-inflammatory and antitussive effects, mainly used in the clinical treatment of acute and chronic bronchitis and other respiratory diseases, with high efficiency and low toxicity characteristics. Pre-research group under the auspices of the National Natural Science Foundation of China (NO: 30371766,30572355), confirmed the loquat active parts of triterpenoid acid (triterpene Acids of Loquat Leaf, TAL). TAL has significant anti-inflammatory and immunomodulatory effects, preliminary studies also confirmed the TAL role in the prevention and treatment of chronic bronchitis. Given the chronic bronchitis and pulmonary fibrosis are chronic inflammation, and chronic airway inflammation in the early stages of pulmonary fibrosis, TAL has a preventive effect on chronic bronchitis, pulmonary fibrosis, whether also has a preventive effect? ??Paper, endotracheal intubation bleomycin copy Wistar rat model of pulmonary fibrosis selection of prednisone acetate tablets as a positive control in this study based on loquat triterpenoid acid on bleomycin induced pulmonary fibrosis in its part of the mechanism of action. The main contents are summarized as follows: 1.TAL rat lung fibrosis in 120 male Wistar rats were randomly divided into six groups: normal control group, model group, TAL dose group (450,150,50 mg / kg), and prednisone acetate group (3.5 mg / kg), n = 20. In addition to the normal control group, the one-time infusion of the rest of the group intratracheal bleomycin (5 mg / kg) rat model of pulmonary fibrosis and normal control group with saline instead of bleomycin. Treatment group the first two days after the modeling and oral administration of the drugs, 1 times / d, until the end of modeling, the normal control group and model group were treated with the appropriate amount of 0.5% sodium carboxymethyl cellulose solution. TAL dose group can significantly reduce the the rat lungs index lung tissue HA, LN, PIIINP, CIV and Hyp levels were also significantly decreased compared with model group. Histopathological examination and semi-quantitative analysis of the morphology TAL-dose group also showed significant improvement in lung tissue structure, alveolar damage to the structure and degree of alveolar septal thickening was significantly reduced, reduce the infiltration of inflammatory cells, collagen fibers decreased. These results suggest that: TAL significant role in the prevention and treatment of pulmonary fibrosis in rats. Anti-rat lung fibrosis 2.TAL its mechanism compared with model group, TAL can significantly reduce pulmonary fibrosis in rat serum MDA levels, increased SOD activity. RT-PCR results showed that, compared with the model group, TAL significantly reduce the 28 days of pulmonary fibrosis in rats TGF-beta1, CTGF expression was lower in COL-I mRNA expression in lung tissue. These results suggest that TAL prevention and treatment of pulmonary fibrosis in rats may be related to anti-lipid peroxidation, inhibition of TGF-beta1 downregulated the expression of CTGF, thereby inhibiting the synthesis of collagen, reducing the deposition of ECM, to reduce the role of pulmonary fibrosis .
