Abstract


To study the effect of phytic acid, methyl jasmonate and high temperature on growth, heat resistance and the accumulation of secondary metabolites in protocorm-like bodies of Pinellia ternata during suspension culture. At the same time, to discuss the effects of different light quality on growth and development, stress resistance, yield and quality of Pinellia ternata tissue culture seedling.The results are as following:The suitable concentration of elicitor could significantly increase proliferative effect and the accumulation of guanosine, total alkaloids and total free organic acid in protocorm-like bodies of Pinellia ternate. But they were not positively correlated with each other. The accumulation of total alkaloids and total free organic acid was higher under high-concentration methyl jasmonate, but the proliferation of protocorm-like bodies was inhibited. Considering the proliferation of protocorm-like bodies and the synthesis of secondary metabolites, so the medium supplemented with phytic acid0.1%was the suitable concentration.With phytic acid concentration and methyl jasmonate concentration increasing, the content of MDA initially decreased and then increased in PLBs, meanwhile, the content of proline, soluble sugar and soluble protein in PLBs initially increased and then decreased. With phytic acid concentration increasing, pH of the culture medium when harvested and activities of SOD and POD initially increased and then decreased. But with methyl jasmonate concentration increasing, pH of the culture medium when harvested and activities of SOD and POD ascended at all times. The results show that elicitor could improve the antioxidant ability of PLBs, thus delaying cell aging.In all low-concentration phytic acid and methyl jasmonate treatment, activities of SOD and POD increased, the content of MDA decreased, the content of soluble sugar and soluble protein increased. The results show that low-concentration elicitor could improve the high-temperature stress resistance of PLBs, thus it could increase the proliferation of PLBs. Antioxidant system mainly was induced by methyl jasmonate, and phytic acid mainly influenced accumulation of osmoregulation substance. When methyl jasmonate concentrations were higher than60μmol·L-1or phytic acid concentrations were higher than0.15%, it would aggravate the stress of PLBs and then browning was shortened.Photosynthetic pigments increased under the different light qualities(red light, yellow light, blue light and green light). Among them, yellow light treatment was the better effective and red light treatment took second place. The total chlorophyll content of leaves under blue light was lower than others, but Chla/Chlb was the highest. Net photosynthetic rate, stomatal conductance, transpiration Rate, light compensation point and light saturation point reached maximum under white light. Under red light, intercellular CO2concentration and dark respiration rate reached maximum, but net photosynthetic rate reached the lowest. Apparent quantum yield was higher than others under blue light.Under red light treatment, the activity of SOD and the content of soluble sugar reached maximum, and the plant height was the highest, but the content of soluble protein was the lowest. Under blue light treatment, the content of soluble sugar and MDA reached the lowest, but the activity of POD and the content of soluble protein were the highest. At the same time, the total biomass reached the highest and water content was the lowest. Under white light treatment, the activity of POD and the plant height were the lowest, and the content of MDA reached maximum. Water content reached the highest and the total biomass reached the lowest under yellow light treatment. We also found out that contents of soluble protein in leaves and tubers were not positively correlated with each other. The content of soluble protein accumulated mostly in the tuber of Pinellia ternate.Blue light treatment was better effective for the accumulation of total alkaloids, while green light treatment inhibited the accumulation of total alkaloids, red and yellow light treatment had no obvious effect. The accumulation of total free organic acid increased under the different light qualities(red light, yellow light, blue light and green light). Among them, red light treatment was the well effective, while white light treatment did not favor accumulation of total free organic acid. White light treatment was the better effective for the accumulation of guanosine, and yellow light treatment did not favor it.
