Light is one of the most important ecology factor which plays an important role in growth, quality formation and secondary metabolism of flue-cured tobacco. Using tobacco (Nicotiana tabacum L.) plants as experiment materials, the effects of light intensity and light quality on the growth and nicotine synthesis of flue-cured tobacco were studied in this paper. We want to provide some physiological and ecological base information for the effects of light on the growth and nicotine synthesis of flue-cured tobacco.1Effects of light intensity on the growth and nicotine synthesis of flue-cured tobacco20days after the tobacco seedlings were transplanted in the field, different light intensity were set to discussion effects of light intensity on tobacco growth and nicotine synthesis. The main results of the experiments were showed as follows:Light intensity has great impact on the growth and nicotine synthesis of flue-cured tobacco.Light can regulate growth, development and morphogenesis of plants and effects the accumulation of dry matter. The paper analyzed the effects of light intensity on agronomic character of tobacco. The results showed light intensity had a very great impact on growth indicators of tobacco. With the decline of light intensity, the tobacco stem gradually became thinner, the length, width and the ratio of length to width of middle leaves increased, numbers of availble leaves of the plant decreased, dry weight of every tissues and the biomass gradually reduced, R/S ratio decreased and growth period was extended. The net Photosynthetic rate(Pn), stomatal conductance (Gs), transpiration rate(Tr) also decreased with light intensity reduced. The nicotine content of the root, lower leaf and middle leaf also decreased with light intensity reduced. But light intensity has no distinguished impact on nicotine content of the stem. The content of total amino acid, aspartic acid, arginine, spermine, spermidine, polyamine in the root of flue-cured tobacco decreased with the light gradually increased. The content of methionine and nicotinic acid in the flue-cured tobacco decreased with the light intensity gradually decreased. The results showed that the expression of genes that related to nicotine synthesis were affected by the light intensity. The expression of ADC gene was decreased with the light intensity reduced.2Effects of light quality on the growth and nicotine synthesis of flue-cured tobaccoThe experiments of potted planting were carried out in artificial climate chamber when the eleventh leaf of flue-cured grew out. The different light qualities were obtained from different LED. Using blue (B), green (G), red (R), yellow(Y) purple (P) and white (W) diodes (LEDs), we investigated the effects of different light qualities on the growth and nicotine synthesis of flue-cured tobacco. Set white LED light as the contrast, the main results of the experiments were showed as follows:Light quality has great impact on the growth and niotione nicotine synthesis of flue-cured tobacco.Compared with white LED treatment, red LED treatments promoted the height of the plant, the length and width of leaf, while other LED treatments decreased plant height, leaf length and width. Blue LED treatment promoted stem girth.Blue LED treatments promoted the dry matter weight of the root, decreased the dry matter weight of the stem. However the red LED treatments were not the case.Red LED treatments promoted the biomass of the root, the yellow LED treatments and the green LED treatments were not the case.Red and blue LED treatments increased net photosynthetic rate, yellow and green LED treatments decreased in tobacco leaves.Blue and purple LED treatments promoted the nicotine content of the lower leaves, the red and yellow LED treatments were not the case. Compared with white LED treatments, other LED treatments promoted the nicotine content of the middle leaves.RED and purple LED treatments promoted the nicotine content of the stem.Green, yellow and blue LED treatments are beneficial to accumulation of free amino acid that contained in the root of the flue-cured tobacco. Besides the pueple treatmentS,other treatments are beneficial to the accumulation of aspartic acid. The pueple treatment is beneficial to the accumulation of arginine acid that contained in the root of the flue-cured tobacco.Besides the red treatments, others are beneficial to the accumulation of nicotinic acid that contained in the root of the flue-cured tobacco. The red treatment promted the contet of spermidine, decreased putrescine content that contained in the root of the flue-cured tobacco. The green treatments are not beneficial to the accumulation of spermineYellow and blue LED treatments are not beneficial to the expression of QPT gene. Red and blue LED treatments are not beneficial to the expression of MPO gene, the yellow LED treatment was not the case. Green treatment is beneficial to the expression of PMT gene, the yred and purple LED treatments were not the case. Green treatment also is beneficial to the expression of ADC gene,
