Abstract


Objective Through observing the influence of Bushenhuoxue Recipe on Ovarian cells and T lymphocyte subsets of autoimmune premature ovarian failure mice to explore the molecular immunology mechanism of Bushenhuoxue recipe against autoimmune premature ovarian failure.Method Using the mouse zona pellucida3peptide fragments to establish a mouse model of autoimmune premature ovarian failure.The mice were administered with high-dose,low-dose of Bushenhuoxue Recipe comparing with progynova for four weeks, and then, Optical microscope and micro-capillary cell analysis/counter were used to assay the changes of Ovarian cells and spleen T lymphocytes subsets.Results1.Spleen index of the mouse:Model group, BHF low and high dose groups showed no difference with the blank (P>0.05), but there was significantly different between progynova group and the control group, mice spleen index of other four groups were higher than the control group, progynova group was the highest; Model group had significant difference with progynova group(P<0.01), However, the model group had no significant difference between BHF low and high dose groups(P>0.05); Progynova group had difference with BHF low group(P<0.05),but had no difference with BHF high dose group(P>0.05);There was no difference between BHF low and high dose groups(P>0.05).2. Cytological changes in mouse ovaries:Ovarian follicles of Blank group could see clearly, there was no obvious infiltration of lymphocytes or plasma cells; Model mice ovaries follicles became smaller, and even follicular atresia, folliculars and stromal parts of ovaries had obvious infiltration of lymphocytes and plasma cells;The number of folliculars of progynova group and BHF low group was little higher than the model group,but folliculars and stromal parts of ovaries still had obvious infiltration of lymphocytes and plasma cells;The number of folliculars of BSH high dose group was obviously higher than the model group,and the number of lymphocytes and plasma cells was significantly lower than the model group.3.Changes of CD8+T lymphocytes in spleen:Progynova group and BHF low-and high-dose group had great difference with the blank group (P<0.05), there was no significant difference between the blank and model group(P>0.05),but the model group was higher than the blank group; Progynova group、BHF low-and high-dose group had difference with the model group (P <0.05), were all lower than the model group; BHF low group had difference with progynova group(P<0.05),but there was no difference between BHF high-dose group and progynova group(P>0.05);There was no difference between BHF groups(P>0.05).4.Changes of CD4+T lymphocytes in spleen:Progynova group and BHF low-and high-dose group had great difference with the blank group (P<0.01),but there was no difference between the model group and blank group(P>0.05); Progynova group and BHF low-and high-dose group had great difference with the model group (P<0.01),three forward groups showed a downward trend; Progynova group showed no significant difference with BHF low-and high-dose group(P>0.05), but two later groups showed a downward trend; There was no difference between BHF groups(P>0.05).5.Rratio of CD4+/CD8+T lymphocytes in spleen:Model group had significant difference with the control group (P<0.05), Progynova group and BHF low-and high-dose group had no difference with the blank group (P>0.05); BHF low-and high-dose group had difference with the model group (P<0.05),there was no difference between the model group and pogynova group(P>0.05);Through pairwise comparisons,progynova group and BHF high and low dose group had no significant difference (P>0.05).Conclusion1.BHF can improve the ovarian function of the mice with autoimmune premature ovarian failure.2.BHF can adjust the ratio of CD4+/CD8+T cells in spleens of the mice with autoimmune premature ovarian failure to a normal state,and improve the function of immune system eventually.
