Abstract


ObjectiveTo study the extraction and purification process of Cucurbitacin B fromCucumis melo L,determine the content of Cucurbitacin B in the best condition ofextraction and purification technology, preliminary discuss the preparative liquidchromatography purified Cucurbitacin B.MethodsThe reflux extraction,ultrasonic extraction and the supercritical CO2extraction were taken to study the best extraction conditions of Cucumis meloL,measuring the extraction rate of Cucurbitacin B by high performance liquidchromatography,by uniform design experiment preferred optimum extractionprocess and use statistical methods for analysis of experimental results.Use themacroporous adsorptive resin by the purification Cucurbitacin B and confirm thebest purification process.Preparative high performance liquid chromatographywas used in the further purpification experiment to get higher purity.ResultsWith the supercritical CO2extraction,the Cucurbitacin B content reaches16.46%,higher than the method of reflux extraction11.59%and the method ofultrasonic extraction13.52%.Optimal extraction technology was as follows:extracting pressure of26.0MPa,extracting temperature of55℃,dynamicextracting for3.5h.The purity of Cucurbitacin B could reach79.93%in thecondition of1mL·min-1speed sampling,70%ethanol as eluent,1mL·min-1speedfor elution.Preparative high performance liquidchromatography was used in the further purpification experiment,the purity of Cucurbitacin B could reach above95%.ConclusionsThe result of supercritical CO2extraction in the best condition was betterthan the result of reflux extraction and ultrasonic extraction, it is a new avenuefor the study of Cucumis melo L.Though the purification of the macroporousresin the content of the Cucurbitacin B is elevated. Preparative highperformance liquid chromatography was used in the further purpificationexperiment and get higher purity.
