Abstract


"Aconiti Lateralis Radix Praeparata (ALRP) exhibiting fewer hot characteristics without Zingiberis Rhizoma (ZR)" originated in the medical book Zhengzhi Yaojue. Drug pair of ALRP and ZR is widely applied in clinical treatment of TCM. However, whether is the phenomenon of ALRP exhibiting fewer hot characteristics without ZR objective existence or not? Whether or how to establish a scientific and feasible evaluation methods and indexes? And what is the mechanism of ALRP exhibiting fewer hot characteristics without ZR? Under the guidance of TCM and the Biothemodynamics theory, this paper applied the cold/hot plate differentiating technology and microcalorimetry combined with chemical analysis to systematic study the different change of cold and hot among of ALRP, ZR and the different combination between ALRP and ZR. Meanwhile objectivity of ALRP exhibiting fewer hot characteristics without ZR is studied. The main results are summarized below:1. Study of differences between cold and hot of the complication of ALRP and ZR in different proportionTo study the effeets of the complication of ALRP and ZR in different proportion on normal and cold symptomatic mice based on animal thermotropism behavior by cold/hot plate differentiating technology. The results showed that the remaining rate on hot plate (RR) of Model group had increased significantly (P<0.01) compared with Control group, suggesting an enhancement of thermotropism. Compared with Control group, RR of Control+administration group had decreased significantly (P<0.05or P<0.01). Compared with Model group, RR of Model+administration group had decreased significantly (P<0.05or P<0.01), so that tends to normal levels. The mechanism of ALRP exhibiting fewer hot characteristics without ZR may be a change of the level of energy metabolism such as oxygen consumption volume and ATPase activity and antioxidation such as T-AOC、T-SOD. The results of biological thermodynamic in vitro showed that total metabolic heat (Q) of administration group production in Escherichia coli increased significantly compared with non-administration. And total metabolic heat of the complication of ALRP and ZR in different proportion production group in Escherichia coli increased significantly compared with single-administration.2. Study of differences between cold and hot of the complication of ALRP and ZR in same proportion and different concentration The results showed that dose-response relationship existed between concentration of the complication of ALRP and ZR in same proportion and oxygen consumption volume, ATPase activity, T-AOC、T-SOD and RR (P<0.05or P<0.01). The results of biological thermodynamic in vitro showed the complication of ALRP and ZR in same proportion and different concentration existed dose-dependent with Q in Escherichia coli.3. Study of differences between cold and hot of the complication of ALRP and processed products with ZRCompared with the homologous ALRP group, the complication group had an enhancement of antioxidation and energy metabolism and decreased significantly in RR. The complication of ALRP processed products and ZR decreasing RR, the sequence is that ALRP>P>H>D≈B>Y. The sequence of ALRP processed products affected Q in Escherichia coli is that ALRP>H>D>B>P>Y. And Q of AZ increased significantly compared with ALRP.In conclusion, this paper clarified preliminarily objectivity of ALRP exhibiting fewer hot characteristics without ZR based on thermodynamics. Influencing factors of the change of ’hot’ in ALRP involved processed, compatibility and dose. The mechanism of ALRP exhibiting fewer hot characteristics without ZR may be the level of energy metabolism and antioxidation activity. Meanwhile, this paper provided theoretical basis for building research model of regular pattern of cold and hot compatibility in TCM prescriptions by thermodynamics.
