Objective To investigate the effect of serum from uremia patients onthe expression of miR33a,ABCA1and ABCG1in THP-1macrophages,and to study the effect of different types of blood purification on them.Methods40uremia patients were divided into four groups: before andafter HD(n=23), before and after HDF (n=17).23healthy people wereenrolled.THP-1macrophages were incubated with serum derived fromthese people. Blood lipid levels,hs-CRP were measured and the expressionof miR33a,ABCA1,ABCG1in THP-1macrophage was measured byreal-time PCR as well as western blot.Result With serum treatment,the expression of miR33a in THP-1macrophages was up-regulated (p<0.05) and mRNA, protein level ofABCA1,ABCG1were significantly down-regulated (p<0.05) in uremiagroup as compared to healthy group.The level of serum HDL-c in uremiapatients was significantly lower but hs-CRP was higher than that in healthygroup (p<0.05). In addition, in the pre-and post-HD, miR33a, mRNA andprotein level of ABCA1,ABCG1showed no difference (p>0.05).Incontrast, the mRNA and protein expression of ABCA1,ABCG1inpost-HDF were higher and the miR33a was lower than those in pre-HDF as well as post-HD. Moreover, significant increase of HDL-c(P <0.05) anddecrease of hs-CRP (P <0.05) were showed in post-HDF compared withpre-HDF and post-HD, but HD appeared no significant change in thesesubjects.Conclusion In comparison with HD,HDF can down-regulated theexpression of miR33a,then up-regulated the mRNA and protein expressionof ABCA1,ABCG1in macrophages and elevated the level of serum HDL-c,decrease hs-CRP, indicating the effect of anti-atherosclerosis.
