Abstract


ObjectiveTo understand the mechanism by which NCTD attenuates renal fibrosis,the effect of NCTD on inhibition of renal fibrosis in UUO rats was investigated.we also investigate the expressions of FN、collagen Ⅰ、 α-SMA、E-cadherin in human renal proximal tubular epithelial cell line treated with TGF-β1and norcantharidin.We both investigate the expressions of PP2Ac.Methods1.Human kidney proximal tubular epithelial cell line(HK-2)cells were incubated with serum-free DMEM for24hours to synchronize the cell growth, then the cells were divided into3groups:control group(only FBS-free DMEM), TGF-β1(treated with TGF-β15ng/ml)and NCTD group (treated with TGF-β1and NCTD). The cells were collected to extracte total RNA and protein at incubation.2.Renal lesions and the expression of FN、Col-Ⅰ、E-cadherin anda-SMA、PP2Acwere detected by irnrnunohistochemistry and Western blot.RT-PCR examed the expression of FN、Col-Ⅰ、E-cadherin anda-SMA、PP2Ac mRNA,respectively.Results1. Compared with sham group,in UUO group,the expressions of FN、 Col-I increased, simultaneously, the expressions of a-SMA increased and the expressions of E-cadherin decreased in operated kidneys. However,after treatment of NCTD,these changes were partly alleviated in a dose dependent manner.2. Compared with sham group,in UUO group,the expressions of PP2Ac increased,while NCTD downregulated its expressions in a dose-dependent manner.3.Compared with control group,TGF-β1group upregulated the expression of FN、Col-Ⅰ,and promote EMT process,while NCTD group could downregulated the expression of FN、Col-Ⅰ and retard EMT process.4.Compared with control group,TGF-β1group upregulated the expression of PP2Ac,while NCTD group could downregulated its expression.Conclusion(1) NCTD alleviates the tubulointerstitial lesions and EMT process.(2) NCTD inhibited the expressions of PP2Ac in UUO rats and in human renal proximal tubular epithelial cell line (HK-2) induced by TGF-β1.(3)The antifibrotic effect of NCTD may be related to its target-inhibition of PP2A.
