OBJECTIVE: Some researchs found that stromal cell derived factor-1(SDF-1)and chemokine receptor4(CXCR4) plays an important role in the development ofa variety of tumor invasion and metastasis, this paper aims to discussion theexpression and clinical significance of SDF-1and CXCR4two proteins in cervicalcarcinoma.METHOD: Immunohistochemical staining method was used to detect expressionof SDF-1and CXCR4protein in80cases of cervical carcinoma,20cases ofnormal cervical tissue, then analysis and comparison of the relationship betweenSDF-1and CXCR4in age, pathological grade,clinical stage, and lymph nodemetastasis of cervical carcinoma patients, and to research the relationship betweenthe occurrence and development of cervical carcinoma and the expression of SDF-1and CXCR4,and to explore the correlation between the two proteins, for providinga new method of molecular targeted therapies for the diagnosis and treatment ofcervical carcinoma.RESULT:1. Expression of SDF-1protein in normal cervical tissue and cervicalcarcinoma,and its correlation with clinicopathological factors of cervical carcinoma.The positive rate of SDF-1protein in normal cervical tissue was5%(1/20)andthe positive rate of SDF-1protein in cervical carcinoma was55%(44/80). thedifference between the two positive rates was statistically significant(P<0.05).The positive rate of the expression of SDF-1protein in clinical stage≥Ⅱa wassignificantly increaser than the stage <Ⅱa(P<0.05); the SDF-1positive rate inlymph node metastasis was significant increaser than those without lymph nodemetastasis(P<0.05); the SDF-1positive rate in histological grade Ⅰ, Ⅱ, Ⅲwasstatistically different (P <0.05), the positive rate of SDF-1in pathological grade Ⅲgroup was significantly increaser than in pathological gradeⅠ, Ⅱ group (P<0.01, P <0.01), but the SDF-1positive rate in histological grade Ⅰ, Ⅱ groupwas not statistically different (P>0.05). The positive rate of SDF-1protein expression in cervical carcinoma had norelationship with patients about independent of age (P>0.05), tumor size (P>0.05),organization type (P>0.05), the difference was not statistically different.2. Expression of CXCR4protein in normal cervical tissue and cervical carcinoma,and its correlation with clinicopathological factors of cervical carcinoma.The positive rate of CXCR4protein in normal cervical tissue was10%(2/20)and the positive rate of CXCR4protein in cervical carcinoma was52.5%(42/80).the difference between the two positive rates was statistically significant(P<0.05).The positive rate of the expression of CXCR4in lymph node metastasis wassignificant increaser than those without lymph node metastasis(P<0.05); thepositive rate of the expression of CXCR4in clinical stage≥Ⅱ awas significantlyincreaser than the stage <Ⅱa (P<0.05);the positive rate of CXCR4in primarytumor size≥4cm was significantly increaser than those in the primary tumor size<4cm (P <0.05); the positive rate of CXCR4in histological grade Ⅰ, Ⅱ, Ⅲ wasstatistically different (P <0.05), the positive rate of CXCR4in pathological grade Ⅲgroup was significantly increaser than in pathological gradeⅠ, Ⅱ group (P<0.01, P <0.01), but the CXCR4positive rate in histological grade Ⅰ, Ⅱ groupwas not statistically different (P>0.05).The positive rate of CXCR4in cervical carcinoma had no relationship withpatients about in age (P>0.05), organization type(P>0.05), the difference wasnot statistically different.3. The correlation of the expression of SDF-1, CXCR4protein in cervicalcarcinomaThe expression of SDF-1and CXCR4in cervical carcinoma were increased,there was a positive correlation (R=0.473).CONCLUSION: The high expression level of the chemokine CXCR4and itsligand SDF-1protein was involved in the growth, proliferation, invasion andmetastasis of cervical carcinoma, their positive correlation might have somerelationship with the occurrence and development of cervical carcinoma.
