Abstract


Background: Cervical cancer is a common female reproductive system tumor.The incident of cervical cancer is just followed that of breast cancer which is a seriousthreat to the physical health of women. Because the typical symptoms of cervical cancerare not always clear, most of the patients are hardly aware of the problems. Cervicalcancer early diagnosis is a very important to improve the survival rate. Currently, themain methods of Cervical cancer early diagnosis consist of vaginal smears, colposcopyand biopsy pathology. Vaginal smears and colposcopy could make an error to diagnosisof cervical cancer. Biopsy pathology, whose survival rate is very high, could injury tothe patients. So looking for a non-invasive, high precision method to diagnose cervicalcancer is very necessary. With the rapid development of molecular biology, tumormarkers have been paid more and more attention. Evaluation of screening and prognosisof cancer has important clinical value. The emergence of the morbid proteomics hasresulted in a big step of the research for tumor makers. The main form is to use a varietyof effective methods to separate and purify some specific proteins from specific diseases.Then the specific proteins compared with healthy people are analyzed by MS.As we all know, protein as the main bearer of the body functions is involved intumor development. Glycosylation and phosphorylation are two common proteinpost-translational modifications, affecting the all aspects of the body activities, such ascell proliferation, cell differentiation, cell signaling and so on. The abnormalglycosylation and phosphorylation is a key step to the development of tumor. Manyextracellular glycoprotein sugar chains containing sialic acid are proved to be related tothe development of tumor. Take advantage of proteomics analysis to glycosylation andphosphorylation of proteins, research the development of them in diseases are veryimportant step in clinic.Objective: Present experiments using titanium dioxide as the enriched material, and sialic acid glycopeptides and phosphopeptides were collected separately from thesaliva of healthy women and patients with cervical cancer. And then take advantage of“Click TE-Cys” which was synthesized by DICP to separate glycopeptides andphosphopeptides. In the last glycopeptides and phosphopeptides were analyzed by MS.Methods: Take advantage of the standard samples to establish a trypsin digestionand enrichment and separation of metal oxide and "Click the TE-Cys material, afterjoint MS detection method. Saliva from10tumor patients and10normal healthywomen were used the above method to detection and compared with each other.Results:1, we established a method of titanium dioxide enriched salivaphosphopeptides and sialic acid glycopeptides, synthetic materials "Click TE-Cys"separating two polypeptides, in the last take advantage of MS to detection.2, using the above method, the N glycopeptides and M phosphopeptideswere found in the saliva of the patients with cervical cancer, and the N glycopeptidesand M phosphopeptides were found in the saliva of healthy women.Conclusion: phosphopeptides in the saliva of patients with cervical cancer andglycopeptides compared with healthy people. We found that there are many differentpeptides in the two kinds of people. We think that this method will improve thedetection accuracy of cervical cancer.
