Abstract


ObjectiveObserving ray cartilage polysaccharides on mouse melanoma xenografttumor angiogenesis mimicry of the temporal and spatial variation, and CD31(platelet-endothelial cell adhesion molecule, PECAM-1), MMP-2(Matrix metallo-proteinase-2), HMB45(Human Melanoma Black)cytokine expression, and toprovide a theoretical basis for the screening of antagonistic drugs.Methods1. Modeling and grouping The B16melanoma cells were seeded in C57mice right armpit, made of mouse melanoma xenograft tumor animal models,tumor, were randomly divided into normal saline group, stingray cartilagepolysaccharides low doses, dose, high dose group and the cyclophosphamidegroup, given different doses of Ray Cartilage Glycosaminoglycans. Accordingto a computer-generated random number table every day in each experimentalgroup extracted a mouse anesthesia overdose killed a day, each group of twotumor samples, lasted12days, received120tumor samples. 2. Stained tumor samples obtained from formalin-fixed, paraffin-embedded,sliced, using immun-ehistochemical staining, CD31, PAS double stainingobserved the exp-ression of cytokines. Thevascular generated contemplated state structure in the tumor tissue of thecentral area and the peripheral area are over time change the first increase,reduce, endothelium-dependent in the tumor tissue of the central area and theperipheral area are reducing, and stingray cartilage polysaccharide dose thegreater the trend more evident. The angiogenesis intends density of states andtheir tumor endothelium-dependent cytokines of CD31, there is a close correla-tion between the expression of MMP-2, HMB45. Compared with the saline group, with the higher concentration of Ray Cartilage Glycosaminoglyca-nseach dose group, angiogenesis proposed state central zone and the perip-heral zone tumor cells CD31expression of MMP-2, HMB45gradually reducedin the center of the tumor, the same endothelium-dependent area and the surro-unding area showed a decreasing trend.ConclusionsRay Cartilage Glycosaminoglycans inhibit B16melanoma xenograft tumorangiogenesis, vasculogenic mimicry and endothelium-dependent vascular the-re are certain laws of time and space changes. CD31, MMP-2, HMB45, exp-ression of vascular generated intends density of states is closely related.
