Abstract


ObjectiveEstablish vitro H2O2-induced rat cataract model and observation teapolyphenols (TP) transcription factor nuclear factor-kB (NF-kB) H2O2-inducedrat lens epithelial cells, and caveolin-1(CAV-1) expression, looking for newways and theoretical basis for the protective effect of tea polyphenols onoxidative damage to the lens and mechanisms for the prevention of oxidativedamage cataract.MethodsCultured in vitro lens technology, placed in a certain concentration of H2O2in MEM medium transparent crystals SD rats, experimental cataract lens model.To set blank control group, H2O2group and tea polyphenols (0.02mg·ml-1,0.2mg·ml-1,2mg·ml-1) treatment group were sampled at different time points (6h,12h,24h,48h)its morphological observation, the use of immunohistochemicalmethods were counted lens epithelial cells, NF-kB, caveolin-1-positive cellsexpressed and NF-kB in the application of the RT-PCR analysis of lensepithelial cells of caveolin-1mRNA inthe expression, Western-blot Western blotanalysis of lens epithelial cells, NF-kB, caveolin-1protein expression changes.ResultsThe blank control rats vitro lens culture within48h no significant changes in the lenses were transparent to the H2O2group of rats in vitro lens culture within48h with H2O2treatment time, the turbidity of the lens gradually worsened. Lensopacity progress of different concentrations of TP experimental groupcompared with H2O2group of slow, the H2O2group were significantly lower thanin the different time periods turbidity. H2O2raised lens epithelial cell expressionof NF-kB, and time-dependent manner (P <0.01), different concentrations of teapolyphenols can inhibit the expression of NF-kB in lens epithelial cells; H2O2down the Caveolin-in lens epithelial cells1expression in a time-dependentmanner (P <0.01), different concentrations of tea polyphenols can inhibit thereduction of Caveolin-1expression in the lens epithelial cells, different times oflens epithelial cells NF-kB, Caveolin-1positive expression rate and thecomparison of the expression levels in each experimental group compared withH2O2group, the differences were statistically significant (P <0.05), the teapolyphenols concentration of0.2mg·ml-1inhibition effect of the most significant(P <0.05).ConclusionNF-kB, Caveolin-1are present in cataract lens epithelial cells in the normallens epithelial cells, with the time extension of H2O2for the expression of NF-kBgradually increase of Caveolin-1expression is gradually reduced. Teapolyphenols can inhibit the reduction of the increase in H2O2-induced lensepithelial cell NF-kB and Caveolin-1expression, tea polyphenols concentrationof0.2mg·ml-1when the best effect.
