Osteoporosis is one of the most common bone diseases in the world andresults from an imbalance of bone cell function. In the process of guided boneregeneration, osteoporosis weakens the bonding strength between material ofmembranes and bone. Naringin,which can stimulate function of osteoblastsand inhibit osteoclast formation, is evidenced to be effective for the treatmentof osteoporosis and bone resorption. In this study, naringin was incorporated inthe electrospun nanomembrane containing poly(ε-caprolactone)(PCL) andpoly(ethylene glycol)-block-poly(ε-caprolactone)(PEG-b-PCL). Thenanomembrane was demonstrated unchanged chemical structure, improvedhydrophilicity, thinner and more uniform nanofiber by Fourier-transforminfrared spectroscopy, contact angle measurement and scanning electronmicroscopy. The nanomembrane also showed faster degradation rate andcontrolled-release of naringin. Osteoblast-nanomembrane interactions werestudied by seeding MC3T3-E1osteoblasts on the nanomembranes,the resultsdemonstrated the controlled-release naringin nanomembranes supportedgreater osteoblast adhesion, proliferation, differentiation, and ECMmineralization. Meanwhile, the response of osteoclasts to nanomembranes wasevaluated in a mouse calvarial critical size defect organ culture model. Theresults indicated the controlled-release naringin nanomembranes suppressedosteoclast formation.
