Clinical and Experimental Researches into Protective Effect of Bicyclol Against Drug-induced Hepatic Injury During Treatment for Severe Psoriasis

Abstract


Objective:Psoriasis is a common chronic, inflammatory dermatosis which easily relapses. During the treatment for psoriasis, medicines like methotrexate and retinoid acid will probably induce hepatic injury in a long term. In this project, the protective effect of bicyclol against drug-induced hepatic injury during treatment for severe psoriasis are researched via both clinical observation and experiment. This may give clues about more optimized clinical treatment for psoriasis on the basis of experiment.Methods:Experiment1:Pick up cases that meet certain criteria, and divide all cases into one treatment group (12cases) and one control group (17cases) by means of a random number table, and are respectively dosed with bicyclol pills (25mg tid) and diammonium glycyrrhizinate pills (150mg bid) after drug-induced hepatic injury appears, until biochemical markers return normal. Observe the recovery rate of hepatic function in both groups.Experiment2:Divide40ICR mice randomly into five groups, which are Group A as blank control group, Group B as model group (dosed with tripterygium), Group C (dosed with high doses of bicyclol), Group D (dosed with low doses of bicyclol), Group C as control group (dosed with diammonium glycyrrhizinate). Group A and B are dosed with carboxymethyl cellulose suspension, and Group C, D, E are dosed with drugs respectively for five days. In the sixth day, Group B, C, D, E are dosed with tripterygium. After24hours, collect blood of mice and kill them in order to collect the liver after dissection. Observe serum markers and apply pathological inspection.Results:Clinical experiment shows that both bicyclol and diammonium glycyrrhizinate are hepatic protective and reducing enzymes (P<0.05), in which the total efficiency are respectively100%and94.12%for bicyclol and diammonium glycyrrhizinate. Thus no significant difference is found for the efficiency of both medicines (P>0.05).Animal experiment shows that, when comparing serum AST, the figure in model group is statistically significantly greater than the figure in blank group (P<0.05). Figures in high-dose-bicyclol group and diammonium glycyrrhizinate group are statistically significantly greater than the figure in model group (P<0.05). And low-dose-bicyclol group is statistically similar to model group (P>0.05). High-dose-bicyclol group is statistically similar to diammonium glycyrrhizinate group (P>0.05). The figure in low-dose-bicyclol group is statistically greater than the figure in diammonium glycyrrhizinate group (P<0.05).Conclusion:Bicyclol is capable of reducing hepatic function which is raised by drug administration, and easing the denaturation and necrosis of hepatocytes, which means it is hepatic protective and effective in reducing enzymes.
