Abstract


Jinghai yellow chicken egg-laying hens, the study for the test material, eggs thiamphenicol, florfenicol and the metabolite florfenicol amine residues by high performance liquid chromatography with fluorescence detection. Through the laying hens were -1 · bw · d Oral Dose thiamphenicol and 50mg · kg -1 · bw · d dose of 20 mg · kg Oral florfenicol, daily administration for 5d, study of the egg white, egg yolk, whole egg thiamphenicol, florfenicol and the metabolite florfenicol amine residues to eliminate the law. The results show that: 1. Established thiamphenicol in egg white, egg yolk and whole egg in the pre-treatment method and HPLC with fluorescence detection method thiamphenicol recoveries in the egg white from 83.43% to 91.23%, days day RSD were less than 5.32% and 10.75%, respectively; thiamphenicol in egg yolk recoveries between 77.46% to 82.55%, the intra-day and inter-day RSD were less than 6.11% and 11.23%; Thiamphenicol neomycin in the whole egg recovery rate from 86.35% to 93.76%, both intra-day and inter-day RSD were less than 6.62% and 10.83%, respectively. Thiamphenicol 0.025mg · L-1 ~ 5.0mg · L-1 range linearly (r = 0.9997, n = 8), detection limit for 3μg · kg -1 , can be meet the requirements of the residues in eggs thiamphenicol. 2 florfenicol and florfenicol amine in egg white, egg yolk and whole egg in the pre-treatment method established HPLC with fluorescence detection method, egg white florfenicol and florfenicol amine recoveries were 81.36 % to 97.87% and from 83.71% to 97.14% between day RSD were less than 6.34% and 5.62%, 11.35% and 11.51%, respectively, lower than day RSD; recovery of florfenicol and florfenicol amine in egg yolk rate from 74.36% to 87.45% and from 79.61% to 97.23%, day RSD were less than 7.37% and 5.82%, 12.52% and 10.36%, respectively, lower than day RSD; whole egg florfenicol and fluorobenzene florfenicol amine recoveries were in the range of 87.41% to 92.29% and from 89.01% to 95.15%, the day RSD were less than 6.73% and 5.82%, 10.48% and 10.52%, respectively, lower than day RSD. Florfenicol a good relationship in the linear range was 0.01 ~ 10.0mg · L-1 (r = 0.9997, n = 9); florfenicol amine in 0.0025mg / L ~~ 2.50 mg · L-1 range a good linear relationship (r = 0.9998, n = 9), the test of florfenicol and florfenicol amine detection limit were 3μg · kg -1 and 1μg · kg -1 to meet the egg florfenicol and florfenicol amine residue testing requirements. 3 to establish the simultaneous determination of thiamphenicol, florfenicol and florfenicol amine in egg white, egg yolk and whole egg pre-processing method and HPLC with fluorescence detection method, egg white thiamphenicol, florfenicol and florfenicol amine recoveries were 83.85% to 92.41%, from 81.21% to 97.64% and from 81.26% to 87.61%, egg yolk thiamphenicol, florfenicol and florfenicol amine recovery rates were in the range of 86.65% to 93.23%, from 87.83% to 92.27% and from 89.56% to 95.86%, whole egg thiamphenicol, florfenicol and florfenicol amine recoveries were 77.56% ~ 91.94 %, from 74.82% to 87.38% and from 78.87% to 97.36%. Thiamphenicol, florfenicol and florfenicol amine detection limits were 3μg · kg -1 , 3μg -1 and 1 μg · kg · kg < sup> -1 . Simultaneous determination to meet the requirements of the eggs thiamphenicol, florfenicol and florfenicol amine residues detected. 4. Laying hens of 20 mg · kg -1 · bw · d dose, continuous 5d, day 1 Oral thiamphenicol residues in eggs thiamphenicol after elimination of . The results show that: with the increase in the number of medication days during the treatment, egg white, egg yolk and whole egg thiamphenicol content gradually increases, reaches a maximum 1d are in withdrawal, its contents were 0.15721mg · kg -1 , 0.18221mg kg -1 , and 0.13732mg kg -1 , and the egg white thiamphenicol content higher than egg yolk Thiamphenicol content of neomycin; during the withdrawal thiamphenicol content, egg white, egg yolk and whole egg in its content gradually decreases with the withdrawal of prolonged days, stopping pre-residual eliminate fast withdrawal late, its residual eliminate slow withdrawal 5d, egg white thiamphenicol residues below the limit of detection (3 μg · kg to -1 ); withdrawal after 7d, the the whole egg thiamphenicol content below the detection limit (3μg · kg -1 ); 8d after withdrawal, egg yolk thiamphenicol content below the detection limit (3 μg · kg -1 ); egg yolk thiamphenicol residues eliminated slowly than the egg white residue, eliminating a long time. 5. Laying hens -1 · bw · d dose to 50mg · kg continuous 5d, once a day orally fluorophenyl eggs after Florfenicol Florfenicol and metabolites florfenicol amine residues in the elimination of the law. The results showed that: during treatment, egg white, egg yolk and whole egg in florfenicol and florfenicol amine content increases gradually with the increase in the number of medication days in the discontinuation of the first day, egg whites, egg yolks and whole egg in fluoride reached the maximum benzene content of florfenicol its contents for 0.66201mg · kg -1 0.75733mg · kg -1 and 0.70227mg kg -1 < / sup>. During the withdrawal, egg white, egg yolks and whole egg florfenicol and florfenicol amine content, with the discontinuation of the number of days of extension of its content gradually decreases withdrawal 5th day florfenicol amine content of the egg white below the limit of detection (1μg · kg -1 ); withdrawal 6d, egg white florfenicol content below the detection limit (3 μg · kg -1 ); withdrawal 9d, whole egg florfenicol content below the limit of detection (3μg · kg -1 ); withdrawal 10d, Florfenicol and Florfenicol amine content in the egg yolk were below the detection limit (3 μg · kg -1 and 1μg · kg -1 ), whole egg florfenicol amine content is below the limit of detection (1μg · kg -1 ). The yolk of florfenicol and florfenicol amine residues than egg white residue eliminated slowly, eliminating a long time. Florfenicol and florfenicol amine sum thiamphenicol and florfenicol () maximum residue limits (MRLs) in the chicken in the standard reference to the European Medicines Evaluation Agency (EMEA) (Thiamphenicol neomycin 50μg · kg -1 florfenicol 100μg · kg -1 ), proposed laying hens to 20mg · kg -1 · bw · d dose continuous 5d, 1 Oral thiamphenicol every day, the withdrawal period (WDT) to 4d; the proposed laying hens 50mg · kg -1 · bw · d dose continuous 5d, once a day orally fluorophenyl florfenicol WDT 7d.
