Abstract


Objective To investigate the therapeutic role of adenovirus vector mediated rat interleukin 10(rIL-10) gene on streptozotocin-induced type 1 diabetes mellitus of KM mice.Methods Culture 293 cell to amplify adenovirus. Seventy-five male KM mice aged from four to five weeks were randomly divided into five groups; every group contained 15 mice. Group 1 is vacuity goup; Group 2 is diabetes mellitus control group. Group 1 is normal control group without any disposal. Group 2-5 received intraperitoneal injection of stz 40mg/kg.d for 5 days in order to induce type 1 diabetic mellitus. When models builded, Group 3 and 4 received intraperitoneal injection of adenovirus vector mediated rIL-10 gene (0.1ml). Group 4 and 5 received regular insulin 6u/kg.d twice per day by neck subcutaneous injection. Mice were fed with a standard diet and lived libitum. Weight and glycemia of all mice were measured once a week. Three weeks later, all mice were executed and pancreata were fixed in formalin for 12-24 hours, then embedded in paraffin, and sectioned. Observe the incidence of diabetic mellitus, the degree of pancreas inflammatory infiltration and the expression of rIL-10 detected by immunohistochemistry. Detected serum IL-10, IL-4, IFN-γ, C-peptide by enzyme linked immunosorbent assay (ELISA).Results Compared with control groups, IL-10 intraperitoneal injection groups have no significal difference in average blood glucose level, body weight increase and pancreas pathology. RIL-10 has a higher expression in immunohistochemistry. C-peptide IFN-γ, IL-4, IL-10 in serum detected by ELISA in experimental groups have no difference.Conclusion These results suggested that adenovirus vector mediated IL-10 cannot signicantly protect streptozotocin(stz) induced type 1 diabetic mellitus of KM mice in early stage.
