OBJECTIVE:To investigate the effect of lipid-bound cysteine mutants of apoA-I on acute gouty arthritis induced by monosodium urate in rats.METHODS:Twenty-five male Wistar rats(140-160g) were divided randomly into 5 groups:saline control group, model group, soybeanlecithin group, rHDL74 group rHDLwt group, and 5 rats in each group. Except the control group were injected saline, right ankle of the other rats were injected Monosodium urate crystals (MSU) to establish gouty arthritis model. Then all the groups rats were injected saline, saline, soy lecithin solution, rHDL74, rHDLwt to intervene through the tail intravenous, respectively. Ankle joints’s welling degree were measured by tape at different time (Oh,2h,4h,6h,8h, 10h). At 1 Oh after measuring, all the rats were anesthetized with ethyl ether, and collected blood from the epicanthal. Then killing them and collecting for leucocyte count, and IL-1βlevels were monitored by Radioimmunoassay. Pathological changes of articular synovium of rats were observed with HE staining.RESULTS:rHDL74 could inhibit joint swelling of rats and reduce levels of IL-1(3 in blood significantly (P<0.05). rHDLwt also could reduce levels of IL-1(3 in blood significantly (P<0.05). Histopathologically, the rHDL74 and rHDLwt could abate synovium proliferation of ankle joint, reduce inflammatory cell infiltration of articlar tissues.CONCLUSION:rHDL74 and rHDLwt have remarkable efficacy in improving symptoms of acute gouty arthritis induced by monosodium urate crystals in rats.
