Abstract


Engineered artificial blood vessels , seed cells (VSMC, EC, Fibroblast) can not form the structure of the natural three-dimensional micro piling up , and can not function normally . In addition to the use stress bioreactor the prepared tubular bionic stent is induced cultivation cells produce natural micro piled structure and function in an important way . Electrostatic spinning and melt spinning can be directly and continuously produced polymer nano / micro fibers , with the winding receiving technology , can be prepared with the circumferential direction of the micro - groove structure biomimetic vascular stent . This study investigated the electrospinning process ( spinning solution concentration , voltage , advance rate , receiving distance , winding speed , etc.) and different melt-spinning process ( melt-spinning temperature , translational velocity , winding speed ) biomimetic tubular scaffold microstructure impact. Combining melt spinning and the manner of using a solvent / non-solvent adhesive by electrostatic spinning is prepared having a two-layer tube wall , the wall having a peripheral tubular to Ordering micro grooves biomimetic vascular stents . Above bracket fixed modified collagen on the cultivation of human umbilical vein smooth muscle cells (VSMCs ) . Fiber winding angles of the VSMCs orientation induced ability using scanning electron microscopy and fluorescence microscopy . The results indicate that the biomimetic scaffold has good biocompatibility, surface circumferential micro- groove structure of VSMCs orientation arranged a significant induction . This has important guiding significance on the preparation of blood for clinical use tube substitute . On the basis of in vitro evaluation of the project also carried a rabbit in vivo transplantation experiments to study the possibility of biomimetic scaffold situ induced angiogenesis . After 1 week in January to remove the tubular stent , its outer layer with rabbit 's own tissues close together ( peel found that the the stent outer layer was pink ) , the cavity has a thickness of about 0.4 mm thrombus formation . Histological observation showed that the circumferential arrangement of VSMCs and fibroblasts attached to the outer surface of the stent .
