Abstract


Chapter 1:The recent development of cyclodextrins and its derivatives have been reviewed,especially its development in drug delivery system.At the same time,the development of the supramolecular system of cyclodextrins and Flavonoids was expatiated.Chapter 2:The formation of the inclusion complex of isoquercitrin(IQ) withβ-cyclodextrin(β-CD) was studied by steady-state fluorescence,UV-vis absorption spectroscopy and induced circular dichroism.The stoichiometric ratio of the three complexes was found to be 1:1 and the stability constants (K) were estimated from spectrofluorometric titrations,as well as the thermodynamic parameters.The effect of pH on the complexation process was also quantitatively assessed.IQ exists in different molecular forms depending on pH andβ-CD was most suitable for inclusion of the neutral form of IQ.The solubility enhancement of isoquercitrin byβ-CD was studied using phase-solubility method.The results showed that it is most suitable for complexation of IQ andβ-CD at lower temperature and acid conditions.β-CD enhanced the solubility of isoquercitrin to 4.64 fold.Chapter 3:The formation of the inclusion complexes of isoquercitrin(IQ) with HP-β-CD and DM-β-CD were studied by steady-state fluorescence、UV-vis absorption spectroscopy and induced circular dichroism.The results showed that the stoichiometry was 1:1.The stability constants were varied at different pH values.With the increase of pH values,the stability of complexes is decreased.The high temperature is unfavorable for the formation of inclusion complexes.The inclusion process is spontaneous,the major forces are form enthalpy change including van der Waals et al.The inclusion with HP-β-CD and DM-β-CD leads to the solubility enhancement of IQ to 7.51 fold and 12.2 fold,respectively. Chapter 4:The formation of the inclusion complexes of quercetin withβ-CD, HP-β-CD and DM-β-CD were studied by steady-state fluorescence and UV-vis absorption spectroscopy.The effect of the concentration of CDs and pH values were also discussed in detail.The results suggested the stability constants for quercetin determined followed the rank order of HP-β-CD＞DM-β-CD＞β-CD.The stability constants were very sensitive to the change of pH values:the three kinds ofβ-cyclodextrin both prefer to encapsulate the neutral molecules than the ion structures.Chapter 5:The formation of the inclusion complex of isoquercitrin(IQ) withβ-CDs were studied by ~1H NMR titrations and molecule modeling technique. The results suggested that IQ inserted intoβ-CDs via the chromone part.
