Neuropeptide Y (Neuropeptide Y, NPY) belongs to the pancreatic polypeptide family, is widely distributed in the animal central nervous system and peripheral nervous system, especially in the telencephalon, preoptic hypothalamus abundant. The study confirmed, NPY is one of the candidate genes for animal feeding and energy metabolism regulation. Within the hypothalamic arcuate nucleus (Arcuate nucleus, ARC; chicken funnel nuclear: Infundibular T nucleus of, IN), paraventricular nucleus (Paraventricular T nucleus of PVN) and lateral (Lateral hypothalamic area, LHA), it is important to feed regulation center. The formation of long-term directional breeding different kinds of production performance of chicken, broiler meat production for the purpose of early growth metabolism, feeding large; while laying performance and slower growth rate, feed than broilers . In this study, from the level of NPY gene transcription, alternative splicing forms and single nucleotide polymorphism (Single nucleotide polymorphisms SNPs) associated with chicken feeding and energy metabolism analysis to clarify the impact of NPY on the chicken food intake and energy metabolism at the molecular level provide a theoretical basis. Test: hypothalamic neuropeptide Y mRNA levels and chicken feeding association analysis RT-PCR using TaqMan probe detection chicken hypothalamic NPY mRNA level, a comparative analysis of NPY mRNA in feed intake, two very different varieties - Hyline and Ross broiler hypothalamic of IN the PVN and LHA transcription level differences. The results show that the weight of the 14-day-old Ross broiler feed intake and 24-hour same-day-old Hyline 1.7-fold and 1.5-fold, respectively, a significant difference among cultivars (P lt; 0.01). Hyline hypothalamus IN NPY mRNA levels in the PVN and LHA was significantly higher than the Ross broiler. The broiler under hypothalamus IN within NPY the mRNA level was significantly higher than the LHA and the PVN, LHA and PVN no significant difference; while laying hens under thalamus IN and the PVN NPY the mRNA level significantly with higher than LHA IN and PVN without significantly with differences. Thus, NPY mRNA expression between the different chicken breeds and organizational differences, just from the transcription level can not directly explain NPY pro-foraging role play in feeding regulation. Trial 2: hypothalamic neuropeptide Y mRNA splicing and chicken feeding association analysis of changes in gene splice sites and splicing allows the gene expression of multiple different protein products, thus affecting its physiological function. The results show: NPY broiler cDNA 3 'end there are two kinds of different forms of splicing fragment sequences appear in the mRNA 3' end terminating short phenomenon. 82bp nucleotide a long sequence 3 'end of the sequence belonging to the non-coding sequence and is not involved in the amino acid sequence encoding the corresponding proteins; the hens cDNA5' end occurred 79bp deletion in the coding region, resulting in changes in the corresponding sites of the encoded protein product, 44 the amino acid sequence deletions, including the signal peptide and the mature peptide sequence, and caused to disappear with the receptor binding sites, thus affecting the secretion and expression activity of NPY. The difference may be speculated that NPY mRNA splicing is partly because of the different cause hens, broiler feed intake and energy metabolism. Test: NPY gene SNPs associated with chicken feeding and fat deposition analysis of this study, PCR-SSCP method to search for SNPs in the AA chicken NPY gene 5'UTR and intron 2 sequences, and combined production, analysis of different genotype and the associated production and carcass traits. Results in amplification the NPY gene intron 2, P12, P13 and P143 primers detected the presence of polymorphic loci, were: SNPP12 (2341 TCT insertion A2342T, T2343C, A2344T, A2345T and C2347G) and SNPp13 -expression of p14 (T2521C, T2597C, T2623C, C2704T, T2776G, T2787C and A2821G), and SNPP12 and SNPP13 ~ 14 haplotype linked genetic correlation analysis showed SNPP12 with 6-week-old AA broilers abdominal fat weight and abdominal The fat was significantly correlated (P lt; 0.05), in which the individual mutation genotype BB abdominal fat deposition is on the increase. The SNP13 ~ 14 and 3 weeks of age, feed intake and 6-week-old liver weight was significantly related (P lt; 0.05) and BB genotype leg muscle rate is significantly higher than the AA genotype (P lt; 0.05). The above results indicate that the mutant genotype BB type of NPY may be favorable genotype energy conversion and fat deposition. In summary, the laying hens NPY mRNA levels were significantly higher than the broiler hens detected NPY mRNA splicing phenomenon exists, its signal peptide and the mature peptide sequence deletions may affect the NPY protein secretion and expression, resulting in bioactive reduce, NPY mRNA splicing may directly affect the chicken feed intake and energy metabolism regulation; of NPY mutation genotype BB type may be favorable genotype energy conversion and fat deposition.
