The vinca alkaloids clinical application is the most widely used and effective natural plant anticancer drugs. Vinca alkaloids in periwinkle very low levels in the body, and the use of chemical synthesis process is too complex, the cost is too high, leading to the contradiction between supply and demand of the vinca alkaloids very prominent. Vinca indole alkaloids in periwinkle secondary metabolites, the external environment conditions of stress (such as seawater stress) can promote alkaloid synthesis Vinca indole alkaloids biological precursor tryptophan, therefore seawater stress The conditions add tryptophan is expected to further improve the vinca alkaloid content. In this experiment, Changchun flower seedlings as test material, under the conditions of the greenhouse study adding exogenous tryptophan roseus seedlings content of secondary metabolites, and ultrastructure of seawater stress conditions clear vinca alkaloids Synthesis and effect adjustment factors provide a theoretical basis and practical foundation. The main results are: (1) under conditions of high concentrations of seawater exogenous tryptophan significantly inhibit roseus seedlings with increasing tryptophan concentration, plant fresh weight, dry weight significantly reduce (P lt; 0.05); add 500 mg · L-1, 750 mg · L-1 tryptophan, periwinkle chlorophyll content, root activity compared with the control (single seawater treatment) significantly reduced; 750 mg · L-tryptophan significantly increased malondialdehyde soluble sugar content. (2) 10% sea water stress conditions, the addition of exogenous tryptophan periwinkle seedlings tryptophan decarboxylase activity had no effect; add 20% and 40% seawater concentration of tryptophan significantly improve roseus seedlings tryptophan decarboxylase activity, add 500 mg · L-1, 750 mg · L-1 tryptophan can significantly promote the accumulation of indole alkaloids. (3) under sea water stress conditions, exogenous tryptophan can significantly improve the periwinkle seedlings alkaloid content of 20% under seawater stress added exogenous tryptophan is more conducive to vinblastine catharanthine vindoline accumulation. (4) 20% sea water stress conditions, the addition of exogenous tryptophan periwinkle vinblastine production significantly increased; 10% and 20% sea water add 250 mg · L-1 tryptophan can significantly improve vincristine yield; 20 % the seawater Add 250 mg · L-1 and 500 mg · L-1 tryptophan, catharanthine vindoline production has been significantly improved; (5) 20% seawater treatment, periwinkle seedlings mitochondrial cytochrome C oxidase activity was significantly reduced, the loose sheet layer thinning, plasmolysis. Add different concentrations of tryptophan, cytochrome C oxidase activity to further reduce the lamellar deformation, loose chloroplast deformation. With the increase in tryptophan concentration gradient, the the chloroplast lamellar structures tend to disintegrate. Plus 750 mg · L-1 tryptophan, even cytoplasmic cavitation chloroplast death.
