Abstract


Soybean mosaic virus (Soybean Mosaic Virus, SMV) disease is a disease in the world, is widely distributed, seriously affecting soybean yield and appearance quality. Domestic and foreign scholars have conducted research on soybean mosaic virus CP protein purpose of this study is to identify the sources collected in recent years by the National Center for Soybean Improvement different part SMV strains in different soybean varieties symptomatic response to study different strains of pathogenic sexual difference, the CP gene by cloning and sequence analysis of to reveal our SMV genetic diversity. The main results are as follows: 1. Identified 28 SMV strain on the 10 soybean varieties symptomatic response. The results showed that the symptoms of type of soybean mosaic virus in soybean plants mosaic and necrosis, some of the plants at the same time the performance of the two symptoms. The symptoms of the same SMV strains in different soybean varieties respond differently to different SMV strains infecting the same symptoms of soybean varieties respond differently. The reaction comes in part from the strains of the same region on soybean varieties may be inconsistent, the reaction may be the same virus strains from different regions of the soybean varieties. Description part of a region popular strains belonging to different strains, different parts of the popular part of the strain may be the same. 28 strains can infect the South agricultural 1138-2 and 8101, anti-infection ability Agriculture, Mingnan and 1138-2 and 8101 is weak. Only five of the 28 strains strains can infect a broad-spectrum anti-source Kefeng 1, indicating that those strains has a strong pathogenic HSBC Branch No. 1, with broad-spectrum resistance. By RT-PCR method for the 28 the SMV strains cDNA, positive clones were obtained using the TA cloning method. Relations from a geographical point of view, the 28 strains tested were from the northern spring soybean area, the Huang-Huai soybean producing areas, and the South soybean producing areas, the phylogenetic tree: 4103 and 4032 from the southern soybean-producing areas, respectively Origin Zhejiang and Shanghai, the phylogenetic tree shows high homology; 4297,4103,4032, SC3, DB630, ZHZH131 phylogenetic tree is closer to the branch. 4297,4103,4032, SC3, from the South, soybean-producing areas, DB630 from the Northeast soybean producing areas, ZHZH131 from Huang-Huai soybean-producing areas. The branches more strains from the south, indicating a definite link in the origin of the strain, and CP sequence homology. However WF63, JN17, HB98, WF64, BJ157, 3257,3331 Huang-Huai soybean producing areas, H150, DB359, DB380, H161, belong to the Northeast spring soybean producing areas, Y049 ,4026,4469-4, 4233 belonged to the southern soybean-producing areas , is closer to the branch in the evolutionary tree. ZHZH131 and ZHZH144 from Henan Province, is a distant branches in the phylogenetic tree isolated; BJ157 and BJ166 from Beijing, also belong to the distant branches in the phylogenetic tree. Description strains may be from the same region in similar branches, may also be in the distant branches. Thus, the amino acid sequence homology between the source of the virus is not clear and needs further study. H150 and Y049 CP, gene homology, close branches, they belong to the SC-11 strain groups. 4026 BJ157 10 varieties resistant and susceptible reaction, H150 and Y049 on the differential host response is the same (at the same time planning to SC-11 strain groups), the high homology between them, phylogenetic tree is close branches; 3257, DB380, H161, 3331 four strains of the evolutionary tree is a closer branch CP gene homology, divided in lines 3257 , DB380, H161 classified SC-11, although the 3331 classified the SC14 strain groups, but the SC-11 and SC-14 only symptoms of a differential host reaction differences. This result demonstrates that the strain on the differential host symptom response and the CP gene homology between the high and low may have some relevance, the CP gene homology between the lines divided the molecular basis. It is noteworthy that strains 4103 and 4032 in 10 varieties of resistant and susceptible reaction quite different, the high homology between them, the phylogenetic tree shows are also very close to the branches. 4026 BJ157 ,4469-4 4233,4103 and 4032, SC3 and ZHZH131, Y049 and H150 strain the homology of twenty-two close, but on the type of reaction in different soybean varieties are not the same. In view of this, the SMV strain CP gene homology with the strains on soybean varieties pathogenic reaction directly related to the existence of further study.
