The Effect of Different Concentration of Urokinase on Stereotactic Aspiration of the Rabbit Intracerebral Hematoma and the Expression of the Heme Oxygenase-1

Objective: In this study, by the effects of different concentrations of urokinase supplemented by stereotactic aspiration rabbit brain hemorrhage , hematoma clear and hematoma around the heme oxygenase -1 (heme oxygenase-1, HO-1) expression , and investigate the urinary kinase for rabbit brain hematoma dissolved optimal concentration . Methods : 50 New Zealand white rabbits ( male or female ) , weighing 2.5 to 3.0 kg . 0.5ml of autologous blood injection the rabbit brain capsule establish cerebral hemorrhage model in New Zealand rabbits were randomly divided into five groups (n = 10 / group ) : a control group ; 2-5 groups for different concentrations of urokinase group , respectively. the the hematoma cavity injection 0.1ml modeling to saline and different concentrations of urokinase concentration were 5000U/ml , 10000U/ml , 30000U/ml 50000U/ml three hours after the completion of the modeling hematoma pumping suction . Each group of five animals were used for detection of rabbit brain hematoma dissolved hematoma volume , five animals is used to detect the rabbit brain water content and brain tissue HO-1 expression . Results : the role of different concentrations of urokinase group and the control group compared with urokinase concentration increase the urokinase role group in rabbit brain hematoma volume dissolved gradually increase the the urokinase role of group hematoma dissolved volume was significantly higher than that of the control group ( P lt; 0.05), which 50000U/ml group dissolved effect . Different concentrations of urokinase role group with the control group compared to rabbit brain water content significantly decreased (P lt; 0.05), with the the urokinase concentration increased 30000U/ml rabbit brain water content decreased , but 50000U/ml group rabbit gradual increase in brain water content , and 10000U/ml , 30000U/ml group compared to a significant difference ( P lt; 0.05 ) ; the urokinase role group HO-1 positive cells expression compared with the control group (P lt reduction ; 0.05). With urokinase concentration increased to 30000U/ml HO -1 expression gradually reduced to 50000U/ml expression gradually increased , but still less than the control group , expression least 30000U/ml group . Conclusion: 0.5ml autologous blood injection model in rabbit brain cerebral hemorrhage , urokinase concentration in 50000U/ml can significantly increase the volume of the rabbit cerebral hematoma dissolved , the rabbit cerebral hemorrhage hematoma around the brain water content increase , HO-1 of The expression is also increased . The rabbit brain hematoma surrounding brain water content and HO-1 positive cells in 30000U/ml group was the lowest .
