Abstract


In the past decades, magnetic nanoparticles (MNPs) have attracted significant attention because of their satisfactory properties and diverse applications. According to the excellent characteristics of the Fe3O4-based multifunctional nanoparticles, it has been to the concern of the researchers.In this study, poly (acrylic acid) (PAA) was used for the synthesis of carboxylic groups modified MNPs (PAAIO) in aqueous solutions with monodispersities and superparamagnetic. Folic acid (FA) and fluorescein isothiocyanate (FITC) fluorophores were successively conjugated with MNPs, which made MNPs be able to target and image in cancer cells. Therefore, high water solubility, monodisperse, superparamagnetic, targeting and intensively fluorescent nanoparticles were prepared. In addition, a kind of magnetic nanoparticles was modified by 4-amino-2,2,6,6-tetramethyl-piperidine-oxyl (4-NH2-TEMPO) to be an Electron Spin Resonance (EPR) used nanoparticles. The functional magnetic nanoparticles were characterized by X-ray diffraction (XRD), dynamic light scattering (DLS), transmission electron microscope (TEM), inductively coupled plasma-atomic emission spectrometer (ICP-AES), and vibrating sample magnetometer (VSM), respectively. The toxicity of the nanoparticles was evaluated and no toxic effect was observed. The imaging and targeting of the nanoparticles were also realized in vitro and in vivo experiments by confocal laser scanning microscopy (CLSM) and Kodak in-vivo FX professional imaging system. The characteristic results indicated that the final products have interesting magnetic, optical and targeting properties for further applications in biological and biomedical fields.
