Objective:to explore the close relationship between the moxibustion’s effect of protecting gastric mucosa and the high expression of Heat Shock Protein 70 (HSP70) and the acupoints’relative specificity through observing the influence of the pretreatment of moxibustion on Zusanli (ST36) and Liangmen (ST21) on the stress-induced ulcer (SU) rats’gastric mucosa injury and comparing it with moxibustion on the non-acupoint.To clarify the internal mechanism of protecting the gastric mucosa of the acupuncture and moxibustion and its close relationship with Foot-Yangming meridian from the level of molecule.At the same time,to provide experimental evidence for the meridian-viscerus-correlati on theory and the clinical acupoint prescription.Methods:Sixty SD rats, both gender (by half), were evenly randomized into four groups:blank group, model group, Zusanli and Liangmen-moxibustion group and non-acupoints group.Preparing SU model using water restraint stress method, evaluating ulcer index (UI) by Guth method,determining the expression of HSP70 and HSP70mRNA of gastric mucosa with immunohistochemical and reverse transcriptase polymerase chain reaction(RT-PCR) technique,detecting the content of endothelin (ET) and prostaglandin E2 (PGE2) of gastric mucosa by radioimmunoassay and malondialdehydein (MDA) by thiobarbiturate technique.Results:1.Compared with blank group, rats’gastric mucosa UI in model group increased, the HSP70 expression and the content of MDA and ET also increased, the GMBF and the content of PGE2 decreased, with remarkable difference (P<0.01 or P<0.05).2. Compared with model group, rats’gastric mucosa UI in Zusanli-Liangmen group decreased, the HSP70 expression became more evident, the content of MDA and ET decreased, the GMBF and the content of PGE2 increased, with remarkable difference (P<0.01 or P<0.05).3. Except the content of ET, other detecting items of Zusanli-Liangmen group were remarkable different with those of non-acupoints group (P<0.01 or P<0.05).Conclusion:Moxibustion pretreatment on Zusanli and Liangmen could protect gastric mucosa. Its mechanism includes inducing gastric mucosa HSP70 and HSP70mRNA high expression, increasing GMBF, promoting PGE2’release and restraining MDA and ET producing, with the relative specificity of acupoints.
