Detection of k-ras Gene Point Mutation and DPC4 Gene Mutation in Peripheral Blood of Pancreatic Adenocarcinoma

Abstract


Objective:To evaluate the clinical significance of detecting K-Ras gene point mutation and DPC4 gene mutation in peripheral blood of patients with pancreatic cancer. Methods: Select 12 patients with pancreatic cancer and 17 patients with non-pancreatic carcinoma in the same period, we studied K-ras gene 12 point mutation by PCR-MASA (mutant allele specific amplification) and studied the loss and mutation of DPC4 by ordinal PCR and PCR-DSM in peripheral blood. The data were analysed withχ2 test or Fisher’s exact test. Results:The rate of the 12th codon of K-ras gene point mutation and the one of loss of DPC4 respectively was 75%(9/12) and 25% in pancreatic cancer group, and that respectively was 5.88%(1/17) and 0 in non-pancreatic carcinoma group; The sensitivity and specificity of K-ras gene mutation in diagnosis of pancreatic carcinoma were 75.0% and 94.12%. We didn’t find DPC4 gene point mutation by direct PCR-DNA sequence. DPC4 gene mutation and k-ras gene mutation were not associated with patient’s sex and age, tumor size, location and TNM stage in pancreatic cancer group.(P> 0.05). Conclusions:(1) There is no correlation between DPC4 gene mutation k-ras gene mutation and the patient’s sex, age, tumor size, tumor location and TNM stage in pancreatic cancer group. (2) Detection of both K-ras gene and DPC4 gene mutation in peripheral blood may be a diagnostic method for ditiguish pancreatic benign lesions from malignant ones (3) The detection of DPC4 gene mutation may not be a candidate combining with K-ras gene mutation which can be used to diagnose pancreatic cancer.
