Abstract


In this study, Vicia faba were treated by different nitrobenzene concentration(5 mg/L,10 mg/L,25 mg/L,50 mg/L,100 mg/L). The effects of nitrobenzene on Vicia faba were investigated including the seeds germination, primary roots growth and physiological characters. The genotoxicity of nitrobenzene were evaluated with Vicia faba micronuclei assay, chromosomal aberration and mitotic index.The main results as below:1.The effects of nitrobenzene on Vicia faba seeds germination and primary roots growth Seeds early germination of Vicia faba were promoted by low nitrobenzene concentrations; but the seeding and growth of primary root were inhibited when treat time were longer. The inhibited degree related to the concentration of nitrobenzene.2. The effects of nitrobenzene on Vicia faba root tip cell micronuclei High frequency of Micronuclei was induced by nitrobenzene. Treated by 10mg/L concentration nitrobenzene, Vicia faba root tip cell micronuclei frequency showed significant difference compared to the control(P<0.05), treated by 25mg/L concentration nitrobenzene, Vicia faba root tip cell Micronuclei frequency showed significant difference compared to the control(P<0.01).3. The effects of nitrobenzene on Vicia faba root tip cell Chromosomal aberrationsThe cell chromosomal aberrations increased according to the rise of nitrobenzene concentration. Treated by 5mg/L concentration nitrobenzene, chromosomal aberrations frequency showed significant difference compared to the control(P<0.01), treated by 50mg/L concentration nitrobenzene, chromosomal aberrations frequency reached peak value of 8.88‰.4. The effects of nitrobenzene on Vicia faba root tip cell mitotic index Vicia faba root tip cell mitotic index increased following nitrobenzene concentration when plants treated by 5-50mg/L concentrations, while fell down when plants treated by 100mg/L concentrations. The mitotic of Vicia faba root tip cell were inhibited by 100mg/L nitrobenzene concentration.5. Antagonism about antioxidant vitaminC and toxicity of nitrobenzene10mg/L vitaminC inhibited the micronuclei and chromosomal aberrations which induced by nitrobenzene effectively. It implied that the toxicity of nitrobenzene on Vicia faba arose by free radical oxygen because vitaminC can reduce peroxide degree and super oxygen anion level.
