Optimization of the Extraction for the Total Saponins in Albizia Julibrissin and the Initial Pharmacodynamics Study of the Extracts’ Antiangiogenesis Ability

Abstract


The strategy of anti-angiogenesis for anti-tumors had been affirmed. The new drug development around the inhibition of endothelial cells had received much attention. This study extracted the total saponins of Albizia julibrissin, which had ability of inhibiting endothelial cells, optimized the parameters, and initially discussed the extracts’pharmacodynamics on the HMEC-1 cell, with the extraction rate of the extracts, the content of the total saponins and the inhibition on HMEC-1 cells as evaluation.The spectrophotometric quantitative method was established to determine the content of total saponins of Albizia julibrissin. Taking oleanolic acid as the standard, after being incubated the OD value of sample solution was determined at 560 nm. This method was simple, and the results were stabe and reliable. The recovery was 107.86%.After the optimization experiments, the extraction and separation parameters were determined: liquid-solid ratio of 60, ethanol concentration of 70%, extraction times of 2, and extraction time of 2.5. The D101 macroporous resin chromatography with diameter height ratio of 1:5 was used to separate the total saponins. After static adsorpting for 2 h, 3BV of 0, 30%, 70%, 95% ethanols were used to elute the total saponins with elution flow rate of 1.5 mL/min. The freeze-dried powders of extracts were the total saponins of the Albizia julibrissin. The process route was stable and reliable. The extraction rate of the extracts from the Albizia julibrissin was nearly 3%, increased 2-fold comparing the process without the optimization, and the content of the total saponins was nearly 63.17 %.Initial vitro-pharmacodynamic study showed that the extracts possessed the significant inhibitory activity against the HMEC-1, with the IC50 of 1.5μg/mL. The activity increased 20-fold comparing with the Albizia decoction, 7-fold comparing with the extracts from the process without the optimization. FCM analysis showed that the cell cycle was blocked at sub G0/G1 phase. With the increasing of drug concentration and dosing time, the cell death phenomenon occurred.This study established the extraction process of the total saponins of the Albizia julibrissin extraction with the extraction rate of the extracts, the content of the total saponins and the inhibition on HMEC-1 cells as evaluation, got the extracts with the effect of inhibiting the endothelial cells. This extracts could declare the fifth new drug of traditional Chinese medicine as an effective part of Albizia julibrissin, and further developed into a new anti-tumor angiogenesis.
