Abstract


Salinization of soil is one of the major abiotic-stress that influences agricultural production and environment.With the growing of world’s Population and the decreasing of field,how to develop and utilize salinized-soil becomes more and more important.About 300 million mu of China salinization of soil,accounting for about 25% of fertile soil.With the dramatic increase in our population,the rapid industrial development and massive use of land for housing,so that a sharp decline in China’s fertile soil,and unreasonable soil irrigation,and coastal beach abound further deterioration of the situation of China’s fertile soil.Based on 29 accessions of Brassica napus germplasm introduced from Nantong Institute of Agricultural Science,the aim of this study to screen elite Brassica napus germplasm of salt-tolerance by evaluation of salt-tolerant capacity at two stages of germination and seedling and research some seedling’s physiological factors related to salt-tolerance.the effect of NaCl stress on germination characteristics and physiological and biochemical characteristics at seedling stage were studied in this article to provide theoretical basis for selecting and breeding salt-tolerance varieties of Brassica napus and cultivating Brassica napus in saline land.The results as follows:(1) According to different Brassica napus germplasm of agronomic characters, using multivariate analysis of variance,factor analysis and cluster analysis,evaluation of varieties of Brassica salt tolerance.29 varieties of Brassica can be divided into high-resistant materials 06x-15,6013,6023,6031,etc.5 varieties;salt material 6046,6045,6025,05x-27,etc.10 varieties; in resistant materials 05x-1,6078,6051, 6009,etc.10 varieties and salt-sensitive materials 6041,6042,6077 and 05 x-10.(2) Seedling phenotype to index of salt harm and average of fallen leaves of ten plants can test materials for classification of salt-tolerance,Conveniently;The relationship between the physiological and biochemical indicators showed that malondialdehyde,proline content,PSII photochemical efficiency and relative conductivity are among the ones were not significant or highly significant correlation, and so the use of MDA,proline content,PSII photochemical efficiency and relative conductivity in the identification of any one indicator of salt tolerance of Brassica,can not just look at physiological changes,We should be combined with other indicators and the determination of phenotypic identification at seedling stage.In order to accurately salt-tolerance of Brassica napus.We evaluate the various indicators of our comprehensive evaluation,to overcome the lack of a single index.(3)Brassica napus germplasm 6013 as the excellent material,we extracted the root protein at 5 leaf stage.Some satisfied pictures were obtained through optimizing many key procedures,such as sample preparation,sample dissolution,IPG stripe,IEF electrophoresis,SDS-PAGE.established for Brassica napus root proteomics technology platform.It can support sufficient assistance in Brassica napus root proteomics.
