Abstract


In order to develop organophosphorus hydrolase biosensors, we use repetition silver mirror reaction of silver quasi-reference electrode and the conductive strips, prepared by anodic oxidation of silver / silver chloride reference electrode potential, the work explores the carbon paste electrode production process; research immobilized materials, specifications, auxiliary protein concentration on the effect of immobilization; optimization of temperature, pH and other sensor parameters to determine the optimum conditions the enzyme biosensor, and test the consistency of the sensor, and a stable detection limit sex. The main contents include three parts, the first part: thin base electrode preparation; Part II: One piece immobilized enzyme preparation; Part III: enzyme biosensor performance testing. Obtained the following research results: (1) Silver mirror reaction matrix is ??the best PVC insulating tape, the reaction reagent composition was 2% silver nitrate, 5% sodium hydroxide, 2% aqueous ammonia, 10% glucose; optimal reaction temperature is 45 ℃; light silver mirror reaction in favor, but avoid direct sunlight; optimum reaction time was 10min; matrix is ??soaked in lye on the advance in silver mirror reaction effect was not significant. All reagents should now with the current silver ammonia solution in the preparation process must not add excess ammonia. (2) Silver quasi-reference electrode optimum preparation process: silver mirror reaction nine times, an area of ??98mm2; substrate concentration, temperature, pH and other factors on the silver quasi-reference electrode polarization resistance ability was not significant. Silver / silver chloride reference electrode quasi-optimal preparation as follows: dilute hydrochloric acid concentration 0.1mol / L, anodic oxidation time 3min; electrode area of ??confrontation polarization ability to influence is not significant. Preparation of dimethyl silicone oil as a binder carbon paste electrode, which is better than liquid paraffin; article identified by the M high-purity graphite powder: M 7:3 dimethicone work preparing carbon paste electrode. Determined by cyclic voltammetry scanning whichever silver reference electrode thin difference between the electrodes is less than 10% (3) the aperture of enzyme immobilization carriers that affect the immobilized enzyme film the main factors, the optimum composition of the fixing material and reagents: diameter 0.8μm, cellulose nitrate film thickness of 135μm 10% bovine serum albumin. (4) in the initial response of the sensor when the signal fluctuations, the best acquisition time 45s; hydrolysis time 3min, a temperature of 60 ℃, pH 8.0, the amount of enzyme 280U; enzyme sensor detects the difference was 7.5% ; enzyme tablets be stored at 4 ℃, 24h after the loss of activity both greater than 50%, so the enzyme tablets prepared in this experiment for single use only. The optimum test conditions, the minimum detection limit of the sensor was 0.001 g / L, pesticide residue standard curve was exponential distribution, exponential equation: y = 3.277x-1.4486, R2 = 0.9828. To restore the signal of the sensor for object detection, the signal to be several times the oxidized signal, but susceptible to affect the background signal, the detection limit is relatively high, and the associated linear relation can be presented, large differences between the data, requires further study.
